摘 要:研究刀豆氨酸的提取及2,4-二硝基氟苯与刀豆氨酸在pH9.5硼砂缓冲溶液中反应的最佳条件,进而优化测定刀豆氨酸的方法。运用分光光度法测定2,4-二硝基氟苯与刀豆氨酸的络合物在571nm处的吸光值。结果表明,刀豆氨酸水提取法效果最好,DNFB-乙腈的用量为30μL时,刀豆氨酸与DNFB反应的络合物在571nm处有最大吸收值。在30min内稳定以后该络合物的OD值随着时间的增长而增加,对应线性回归方程为y=0.0083x-0.052,相关系数R2=0.9986。The extraction of L-canavanine and the best reaction conditions of L--canavanine and 2, 4-dinitrofluorobenzene in pH 9.5 borax buffer were studied in this paper, which could optimize the method of determination. The OD value of L-canavanine and 2,4-dinitrofluorobenzene' s complex in 571 nm was determined by spectrophotometry. The results showed that the best extraction method of L-canavanine was water extraction. When the volume of DNFB-acetonitrile was 30 p,L, the complex reaction absorption value of L-canavanine and DNFB in 571 nm was maximum. The OD value of the complex increased with the extension of time after stability within 30 minutes. Corresponding linear regression equation was y=0.0083x-0.052; Correlation coefficient was R2=0.9986.
