摘 要:目的了解泸州本地质粒介导的耐多药大肠埃希菌的CTX M、CTX M 3基因分布情况。方法用K-B法进行耐药菌株筛选,PCR扩增CTX M、CTX M 3基因,琼脂糖凝胶电泳检测基因大小,阳性产物外送测碱基序列。结果 71株大肠埃希菌中,有一株死亡。全敏6株,占8.57%,单耐16株,占22.86%,双耐18株,占26.71%,耐多药30株,占42.86%。耐多药质粒中的阳性结果经DNA测序仪检测基因序列,与已知的基因相比较,CTX M有23株,占76.67%,CTX M 3有21株,占70%。两种基因均含有的有12株,占40%。结论泸州地区耐多药大肠埃希菌存在质粒介导的CTX M、CTX M 3基因,耐多药的大肠埃希菌对第三代头孢菌素中的头孢唑肟敏感,对亚胺培南,美罗培南较敏感。Objective To explore the distribution of plasmid-mediated CTX-M and CTX-M-3 in multi-drug resistant Escherichia Coli (E. coli) in the area of Luzhou City. Methods The bacterial strain with drug-resistance were screened by K-B method; CTX-M and CTX-M-3 genes were amplified by PCR and the gene size was detected by agarose gel electrophoresis; the base sequence of posi- tive products was determined in other research institution. Results Out of the 71 strains of E.coli, 1 strain died, 6 were fully sensitive to antibiotic (8.57%), 16 were single-drug resistant (22.86%), 18 were double-drug resistant (26.71%) and 30 were multi-drag resistant (42.86%); the gene sequence of positive multi-drug resistant plasmids were detected by DNA, which revealed that 23 strains were of CTX-M (76.67%) while 21 strains of CTX-M-3 (70%), and 12 strains were of both gens (40%). Conclusions Multi-drug resistant E.coli in the area of Luzhou City are of plasmid-mediated CTX-M and CTX-M-3; multi-drug resistant E. coli are sensitive to ceftizomine (CFX) of the third generation cephalosporin, and sensitive to imipenem (IPM) and meropnem (MPM) as well.
