针刺对感光细胞凋亡大鼠视网膜形态学的影响

摘　　要：目的:观察针刺对感光细胞凋亡大鼠视网膜形态学的影响,明确针刺对感光细胞凋亡的干预作用。方法:N-甲基-N-亚硝基脲以50mg/kg腹腔注射于8周龄SD大鼠,造成感光细胞凋亡动物模型,将大鼠分为4组:针刺组,正常组,阴性对照组,维生素A治疗组。针刺组每日选取新明,睛明,承泣,球后,肝俞,肾俞针刺治疗,各穴位平补平泻,留针30min,连续治疗14天;维生素A组每日给予维生素A治疗;正常组,阴性对照组不做特殊处理。各组14天后用HE染色观察形态学改变,用TUNEL检测凋亡结果。结果:治疗后针刺组与正常对照组比较,平均光密度增加,提示感光细胞发生凋亡,差异有统计学意义(P<0.05);与阴性对照组及维A对照组比较,平均光密度显著降低,提示感光细胞凋亡减少,差异有统计学意义(P<0.05)。结论:针刺能显著抑制感光细胞凋亡大鼠视网膜形态学的损害,提示针刺是治疗感光细胞凋亡的有效方法。Objective：To observe the effect of acupuncture on photoreceptor cells apoptosis, especially on retinal pigment degeneration intervention. Methods ： MUN was injected with 50mg/kg to abdominal cavity of SD rats with ages of 8 weeks to induce animal models of photoreceptor cells apoptosis. The rats were divided into four groups ： the acupuncture group, normal group, negative control group, vitamin A treatment group. The acupuncture group was given daily acupuncture treatment with Jinming, Chenqi, Xinming, Qiuhou, Ganshu,Shenshu for 30rain,continuously for 14 days;Vitamin A group was given daily vitamin A treatment;normal group and model group without special treatment. HE staining was used to observe morphology change, TUNEL detection was used to test the apoptosis results. Results：Compared with the normal group, the average optical density increased in acupuncture group,indicating photoreceptor apoptosis （P 〈 0. 05）;acupuncture group compared with model group and vitamin A treatment group, average optical density was significantly reduced, suggesting decreased photoreceptor apoptosis （ P 〈 0. 05 ）. Conclusion ： Acupuncture may significantly inhibit photoreceptor ceils apoptosis of rat retina morphology, suggesting the acupuncture therapy is an effective method of photographic cell apoptosis.
