银杏叶提取物对急性肝衰竭大鼠肝脏Bcl-2及Bax表达的影响

摘  要:为了观察银杏叶提取物(ginkgo biloba extract,GBE)对由硫代乙酰胺(TAA)所致肝损伤大鼠肝脏的作用及其对Bcl-2、Bax的影响,并探讨其作用机制,试验将大鼠分5组,其中GBE低、中、高剂量组(50,100,200 mg/kg)连续灌服GBE 30 d,1次/d,正常对照组和模型组分别给予等体积的生理盐水;模型组和GBE低、中、高剂量组在第28天用300 mg/kg的TAA腹腔注射,正常对照组腹腔注射等体积的生理盐水,连续3 d,于末次注射后24,48,72小时采血,用试剂盒检测血清中谷草转氨酶(AST)、谷丙转氨酶(ALT)的水平,并用RT-PCR和Western-blot方法检测各组肝脏Bcl-2、BaxmRNA和蛋白水平的表达情况。结果表明:与正常对照组相比,模型组血清中ALT、AST水平明显升高,在肝脏中Bcl-2 mRNA表达水平下调,Bcl-2蛋白水平在48小时时达到最低点,而Bax mRNA和蛋白水平均上调;与模型组相比,GBE组血清中ALT、AST水平明显降低,肝脏中Bcl-2 mRNA和蛋白水平均上调,Bax mRNA和蛋白水平均下调。To study the protection effect of ginkgo biloba extract ( GBE ) on TAA - induced acute liver failure (ALF) and its effect on the Bcl - 2 and Bax, seventy rats were divided into five groups: the control group; the model group ; low - dose GBE group ; middle - dose GBE group and high - dose GBE group~ The rats in the three GBE groups were continuously fed with 50, 100,200 mg/kg for 30days once a day respec- tively, and the other two groups were fed with an equal volume of saline. The rats in the model group and the three GBE groups were continu- ously treated by intraperitoneal injection of TAA(300 mg/kg) at the 28th day for three days, the rats in the control group were also treated by intraperitoneal injection of saline for three days, and then the experimental materials were obtained at 24 h,48 h and 72 h after the last injec- tion. The serum ALT and AST were determined by the kits, Bcl - 2 and Bax were analyzed with RT - PCR. The protein expressions of Bcl - 2 and Bax were analyzed by Western - blot. The results showed that the serum ALT and AST of the mice in the model group were increased sig- nificantly compared with the control group. The serum ALT and AST in mice in the GBE groups were decreased significantly compared with the model group, and the expression of Bcl - 2 mRNA in liver was down - regulated and reached the lowest point at 48 h, but the protein levels and Bax mRNA were up - regulated on average. Gene and protein expression of Bcl - 2 were significantly up - regulated in liver in the GBE groups, but those of Bax were down - regulated.
