摘 要:目的研究原发性肝细胞癌组织中的丙型肝炎病毒(HCV)NS3抗原的表达,探讨其分子生物学机制。方法选取2008年1月-2011年1月医院收治的56例原发性肝细胞癌患者的手术切除标本及其癌旁组织为研究对象,应用原位杂交法和直接酶标免疫组化法分别测定肝癌及癌旁组织中HCV-RNA和HCAg-NS3的表达。结果肝癌组织中HCV-RNA阳性表达表现为细胞核或胞浆内蓝紫色细小颗粒,阳性率为23.21%,高于癌旁组织(3.57%),比较差异有统计学意义(P<0.05);肝癌组织中HCAg-NS3阳性表达表现为胞浆内棕黄色细小颗粒,阳性率为19.64%,高于癌旁组织(3.57%),比较差异有统计学意义(P<0.05);两种方法测定结果共同阳性者9例,共同阴性者41例,检出符合率为89.29%,经配对资料卡方检验,两种方法之间差异无统计学意义。结论HCV NS3Ag主要弥漫分布于肝癌细胞胞浆内,炎性细胞和胆管上皮细胞内未发现HCV NS3Ag表达,说明其与肝细胞癌的发生有密切关系。OBJECTIVE To study the expression of the hepatitis C virus NS3 antigen in primary liver cell cancer tissue, and discuss the molecular biology mechanism. METHODS From Jan 2008 to Jan 2011, the surgical excision liver cancer and cancer adjacent tissue specimens of 56 cases of patients with primary liver cell carcinoma were selected as the research subjects, then the expression of HCV-RNA and HCAg-NS3 in the surgical excision liver cancer and cancer adjacent tissue specimens were detected by in situ hybridization method and directly enzyme immunoassay. RESULTS The positive expression of HCV-RNA in liver cancer tissue was blue purple tiny particles in the nuclei o1: cytoplasm, the positive rate was 23.21%, which was higher than 3.57% of the cancer group, the difference was statistically significant (P〉0.05) ; the positive expression of HCAg-NS3 in liver cancer tissue was tan tiny particles in the cytoplasm, the positive rate was 19.64%, which was higher than 3. 57%of the cancer group, the difference was statistically significant (P〉0.05). The common positive patients of two methods were 9 cases, the common negative were 41 cases, the accordance rate was 89. 29%, the differences were not statistically significant by the matching material chi-square test (P〉0.05). CONCLUSION HCV NS3Ag mainly distributes diffusely in the cytoplasm of liver cancer cells, and the 7expression of HCV NS3Ag has not been detected in inflammatory cells or bile duct epithelial cells, which demonstrates its close relation to the incidence of hepatocellular carcinoma.
