摘 要:目的观察苦荬黄酮对结扎大鼠冠状动脉引起急性心肌缺血的影响。方法采用结扎大鼠冠状动脉建立急性实验性心肌缺血。取成功建立急性实验性心肌缺血大鼠50只,随机为5个试验组,分别为模型对照组、苦荬黄酮大、中、小剂量组[18.0、9.0、4.5(按生药计)g·kg-1]及阳性对照复方丹参片组(2.0 g·kg-1),每组10只。均为一次性灌胃给药,给药体积10 mL·kg-1,模型对照组给予等体积溶媒,用生理信号采集分析系统记录给药后4、5、6 h的胸前多导联心电图,分别取血测定血清肌酸激酶(CPK)及肌酸激酶同工酶(CK-MB)活性,试验结束时取大鼠心脏切片、染色、测量心肌缺血面积,观察苦荬黄酮对结扎大鼠冠状动脉引起急性心肌缺血的保护作用。结果与模型组比较,苦荬黄酮18.0、9.0 g·kg-1能明显降低大鼠血清肌酸激酶(CPK)及肌酸激酶同工酶(CK-MB)活性(P<0.01),可使大鼠心肌缺血面积明显缩小(P<0.05),使升高的∑ST明显降低(P<0.05),限制心肌梗死范围。结论苦荬黄酮对大鼠急性心肌缺血损伤具有明显的保护作用。Objective To study the effect of total flavonoids of Ixeris chinesis ( thumb. ) Nakai { TFIN) on acute experimen- tal myocardial ischemia in rats. Methods Acute myocardial ischemia models were established by coronary artery ligation in rats. 50 rats were randomly divided into five groups, i. e. model control group ( n = 10 ) , high dose TFIN group ( 18.0 g · kg - 1, n = 10 ) , medium dose TFIN group ( 9.0 g · kg - 1, n = 10 ) , low dose TFIN group ( 4.5 g · kg - 1, n = 10 ) and positive control group ( Fu- fangdanshen Tablet,2.0 g · kg-1, n = 10). All groups adopted one - time intragastric administration by the dose of 10 mL · kg-1. The model control group was administered same volume of solvent. Epicardial ECG were recorded 4,5,6 hours after admin- istration by physiological signal acquisition and analysis system. The activity of serum CPK and CK - MB were determined. The protection of TFIN on acute myocardial ischemia which induced by coronary artery ligation was observed through rat heart slices, staining and measurement of myocardial ischemia area. Results Compared with model control group, treatment with TFIN ( 18. 0, 9.0 g · kg-1 ) in rats could obviously decrease the activities of serum CPK and CK- MB(P 〈 0.01 ) , reduce the myocardial is- chemia area( P 〈 0.05 ), decrease the elevating ∑ST( P 〈 0.05 ) and limit the range of myocardial infarction. Conclusion TFIN had significantly protective effect on acute myocardial ischemia in rats.
