摘  要:细叶野牡丹(Melastoma intermedium Dunn)是我国特有的野生花卉,自然界种群数量少、种子繁殖效率低。组织培养方法是其开发利用的有效技术手段。在MS培养基上,细叶野牡丹的茎段腋芽可以直接诱导萌发,其中MS+6-BA1.0 mg.L-1+NAA0.5 mg.L-1的诱导率达到80%。经过MS+6-BA0.5 mg.L-1+NAA0.05 mg.L-1和MS+6-BA0.5 mg.L-1+NAA0.1 mg.L-1继代培养,83%直接在接种芽的基部形成丛生芽,平均每月芽增殖倍数可达3.6。在生根培养1个月后,生根率达100%,其中1/2MS+NAA0.1 mg.L-1根系生长状态最好。幼苗移栽到灭菌的栽培基质V(草炭):V(珍珠岩):V(腐叶土)=1:1:1中,成活率为87%。An experiment was conducted to study the tissue culture and rapid propagation of Melastoma intermedium Dunn, an epidemic species of wild flowers in China. Results showed that the axillary buds of cultured stem segments of M. intermedium were germinated on MS medium. The induction rate on MS medium supplemented with 6-BA 1.0 mg·L^-1 and NAA 0.5 mg·L^-1 was 80%. Eight-three percent of clustered buds were formed at the base of inoculated buds after subculture on MS containing 6-BA 0.5 mg·L^-1 and NAA 0.05 mg·L^-1 or MS containing 6-BA 0.5 mg·L^-1 and NAA 0.1 mg·L^-1. The proliferation coefficient per month was 3.6. The rooting rate reached 100% after rooting culture for one month. The optimal medium for rooting was 1/2MS supplemented with NAA 0.1 mg·L^-1. The rooted seedlings were transplanted into the culture substrate containing peat, perlite and humus soil at a volume ratio of 1 : 1 : 1, and the average survival rate was up to 87%.
