摘 要:目的:通过观察止消通脉宁含药血清对转化生长因子-β1(TGF-β1)诱导人肾小管上皮细胞(HK-2)增殖的影响,探讨治疗糖尿病肾病有效中药复方止消通脉宁在防治肾间质纤维化方面的作用。方法:将HK-2细胞用含10%胎牛血清的DMEM/F12(1∶1)培养基培养;实验分为6组:空白对照组、单纯TGF-β1诱导组(TGF-β110ng/mL)、空白血清对照组(TGF-β110 ng/mL+10%空白血清)、干预1组(TGF-β110 ng/mL+10%低剂量止消通脉宁含药血清)、干预2组(TGF-β110 ng/mL+10%中剂量止消通脉宁含药血清)、干预3组(TGF-β110 ng/mL+10%高剂量止消通脉宁含药血清)。倒置显微镜下观察各组细胞形态,MTT法检测细胞增殖。结果:TGF-β110 ng/mL能显著诱导人肾小管上皮细胞增殖,与空白对照组相比有显著性差异(P<0.05),且其作用随处理时间的延长而增强,但和止消通脉宁含药血清共同作用后,其促细胞增殖作用受到一定抑制(P<0.05)。结论:止消通脉宁能抑制HK-2细胞增殖,在一定程度上具有防治肾间质纤维化的作用。Objective: To explore the mechanism of ZhiXiao TongMaiNing in the prevention and treatment ofrenal interstitial fibrosis by observing the influence of ZhiXiao TongMaiNing on proliferation of human renal tubular epithelial cells HK-2 induced by transforming growth factor-β1 (TGF-β1). Method: HK-2 cells were cultured with DMEM/F 12 (1:1) containing 10% fetal bovine serum and divided into control group, TGF-β1 group(TGF-β1 10 ng/mL), control group of animal serum (TGF-β110 ng/mL +10% animal serum), first intervention group (TGF-β110 ng/mL + 10 % low dose ofZhiXiao TongMaiNing), second intervention group (TGF-β110 ng/mL + 10% middle dose of ZhiXiao TongMaiNing) and third intervention group (TGF-β110 ng/mL + 10 % high dose of ZhiXiao TongMaiNing). HK-2 cell morphology and cell proliferation were observed respectively through the microscope and MTT assay. Result: 10 ng/mL TGF-β1 could induce remarkably proliferation of human renal tubular epithelial cells HK-2, comparison indi- cated notable difference with blank control group (P〈0.05), it can enhance with the growth of time, but cell prolifera- tion was inhibited to a certion extent after combination with ZhiXiao TongMaiNing (P〈0.05). Conclusion: ZhiXiao TongMaiNing can inhibit the proliferation of ilK-2 and prevent renal fibrosis to some extent.
