摘 要:目的:观察南沙参多糖分散片(RPDTS)小鼠急性毒性反应及其对四氯化碳(CCl4)致小鼠肝损伤的保护作用。方法:通过测定小鼠最大给药量考察RPDTS的急性毒性;RPDTS低、中、高剂量(0.75,1.5,3 g.kg-1)灌胃给药7 d后,采用CCl4致小鼠急性肝损伤模型,比色法测定小鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、肝组织匀浆总超氧化物歧化酶(T-SOD)、丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-Px)的含量,观察肝组织病理学改变。结果:小鼠ig RPDTS最大给药量为含生药208 g.kg-1,相当于南沙参临床用量的120倍;RPDTS各剂量组可显著降低急性肝损伤小鼠血清中ALT,AST活性和肝组织中MDA含量(P<0.05),提高肝组织中T-SOD和GSH-Px活性(P<0.05);病理形态学观察显示RPDTS能明显减轻肝脏的病理损伤。结论:RPDTS无急性毒性,对CCl4所致小鼠急性肝损伤具有一定的保护作用。Objective:To evaluate the acute toxicity and the protective effects of Radix Adenophorae Potanini Korsh Polysaccharides dispersible tablets(RPDTS) on acute hepatic injury induced by carbon tetrachloride(CCl4) in mice.Method: The maximum dosage test was used as intragastric administration in mice to observe acute toxicity of RPDTS.After intragastric administration RPDTS(0.75,1.5,3 g · kg-1)in mice for 7 days,the acute liver injury model was induced by CCl4.The levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST) in serum and total superoxide dismutase(T-SOD),malondialdehyde(MDA),glutathione peroxideobjects enzyme(GSH-Px) in liver were determined by kits respectively.Histopathological examination was also carried out.Result: Acute toxicity test showed that the total dose of intragastric administration of RPDTS was 208 g · kg-1(according to Adenophorae tetraphylla) equivalent to 120 times the clinical dose for adults.Acute liver injury test showed that RPDTS could obviously decrease the levels of ALT,AST in serum and MDA in liver(P〈0.05),obviously increase the levels of T-SOD,GSH-Px in liver,the histopathological examination indicated that liver injuries were relieved.Conclusion: RPDTS has no acute toxicity,RPDTS has protective effects on acute liver injuries induced by CCl4 in mice.
