摘 要:以4个野生灵芝菌株为研究材料,以6个外源菌株为参考菌株,采用MEGA4软件对ITS序列进行NJ法聚类分析,构建系统发育树。结果显示4个灵芝菌株ITS序列与另外6个参考菌株对齐序列长度在357~369 bp之间,G/C含量变化为43.4%~57.7%;德昌灵芝1#、德昌灵芝4#和通江灵芝亲缘关系最近,遗传距离为0,德昌灵芝3#与它们的遗传距离为0.0029;与4个野生灵芝菌株亲缘关系由近到远分别是韩芝、红芝、灵芝(中国科学院微生物研究所)、黑灵芝、血芝、云芝;结合4个野生菌株的ITS序列分析结果和形态特征,4个菌株被鉴定为灵芝(Ganoderma lucidum)种类的不同株系。With the bio-software of MEGA4,the ITS sequences of 4 wild Ganoderma lucidum strains and their 6 reference strains were studied by using NJ method to construct the phylogenetic tree.The results showed that the ITS lengths of their strains ranged from 357 to 369 bp,and G/C contents from 43.4 % to 57.7 %.There were the closest relationships among Dechang lingzhi1#,Dechang lingzhi4# and Tongjiang lingzhi,and their strains'genetic distance was merely 0,while there was a genetic distance of 0.0029 between Dechang lingzhi3# and them.Strains,from the close genetic relationships to the farther relationships with the 4 wild Ganoderma lucidum strains,were in order of Hanzhi,Hongzhi,Lingzhi(The Institute of Microbiology,Chinese Academy of Sciences),Heilingzhi,Xiezhi,Yunzhi,respectively.Comparative study of ITS sequences and morphic characteristics indicated that 4 wild strains were subject to different strains of Ganoderma lucidum.
