干扰艾滋病毒抗体酶联免疫法测定的实验性分析

摘　　要：目的探讨血清中混有纤维蛋白、带红细胞血清对艾滋病毒抗体酶联免疫吸附试验测定的影响．方法ELISA常规法（下称常规法）和ELISA改良法（下称改良法）分别对正常对照组500名Anti—HIV（一）的血清，研究组随机抽样1500名艾滋病毒抗体检测样本的血浆、带红细胞血清、无纤维蛋白血清进行配对HIVAb—ELISA检测。结果HIVAb—ELISA结果以S／CO（X±s）表示，常规法测定研究组的血浆、带红细胞血清、无纤维蛋白血清及正常对照组血清的测定值S／CO（x士s）分别为（O．994±0．551）；（0．974±0．532）；（O．777±0．324）；（0．646±0．315），经统计学分析，正常对照组血清与研究组的血浆、带红细胞血清存在差异（P〈0．05），-9研究组的无纤维蛋白血清差异不大（P〉0．05）；改良法测定研究组的血浆、带红细胞血清、无纤维蛋白血清及正常对照组血清的测定值S／CO（X±s）分别为（0．788±0．332）；（0．724±0．315）；（O．715±0．313）；（O．613±0．302），经统计学分析，正常对照组血清与研究组的血浆、带红细胞血清、无纤维蛋白血清无差异（P〉0．05）；结论纤维蛋白、红细胞等干扰艾滋病毒抗体酶联免疫吸附法测定，排除其干扰是必要的。Objective The first to investigate the effect of mixed fibrin（Fib）and RBC in serum on HIVAb enzyme--linked immunosorent assay （ELISA） Methods Traditional ELISA and modified ELISA were applied to measuring serum HIVAb in 500 healthy controls （healthy control group）, 1500 subjets[study group： plasma samples , serum mixed with RBC and nonfibrin serum] Results Traditional ELISA and modified ELISA results of HIVAb were 0. 994±0. 551and 0. 788±0. 332in plasma samples of study group ,0. 974±0. 532 and 0. 724±0. 315 in serum mixed with RBC study group ,0. 777±0. 324 and 0. 715±0. 313 in non--fibrin serum samples of study group ,0. 646±0. 312 and 0. 613±0. 302in serum samples of healthy control group. Statistical analysis of traditional ELISA HIVAb results showed there was significant difference between healthy control group and other groups （except non--fibrin serum of study group）. There was no significant difference in modified ELISA HIVAb results between healthy control group and other groups. Conclusion The serum with Fib and RBC interfere with ELISA HIVAb , and it is necessary to eliminate the disturbance.
