摘 要:目的:探讨丹参多酚酸盐对高浓度同型半胱氨酸诱导的人脐静脉内皮细胞组织因子(TissueFactor)表达的影响。方法:采用ELISA和逆转录-聚合酶链反应(RT-PCR)方法检测不同浓度丹参多酚酸盐(0、0.5、1.0mg/L)在不同作用时间下(2、4、8、12、24、48、72h)对高浓度同型半胱氨酸(50μmol/L)诱导的人脐静脉内皮细胞TF蛋白和mRNA表达的影响。结果:在24h,随着丹参多酚酸盐浓度的增加,同型半胱氨酸诱导的内皮细胞培养液中TF表达呈浓度依赖性下降(P<0.05);当丹参多酚酸盐浓度恒定时(1.0mg/L),随作用时间的延长,同型半胱氨酸诱导的内皮细胞培养液中TF表达水平呈时间依赖性下降(P<0.05)。结论:丹参多酚酸盐可抑制高浓度同型半胱氨酸诱导的内皮细胞TF的表达,并具有剂量和时间依赖性。Objective :To investigate the intervention of Salvianolate on homocysteine induced expression of tissue factor(TF) in umbilical vein endothelial cells. Methods: Different concentrations (0.0.5.1.0 mg/L) of Salvianolate were applied to umbihcal vein endothehal cells under high homocysteine (50p, mol/L) at different time (2.4.8.12.24.48.72h). TF mRNA and TF protein concentration inthe cells were detected by reverse transcription polymerasechain reaction(RT-PCR) and ELISA. Results: Mter intervention with Salvianolate for a certain time period(24 h), both TF protein concentration and mRNA expression level induced by homocystein in endothelial cell culture fluid significantly decreased along with ascending level of Salvianolate. Under a fixed Salvianolate concentration (1.0 mg/L), both TF protein concentration and mRNA expression level in homocystein induced endothelial cell culture fluid decreased significantly over the intervention time. Conclusion: Salvianolate may inhibit the up-regulation of endothelial cell TF expression resulting from high ho- mocysteine in a dose-and time-dependant manner.
