摘  要:目的:探讨连接蛋白43(Cx43)在小鼠胚胎心的时空表达规律及意义。方法:用抗Cx43、抗心肌肌球蛋白重链(MHC)和抗横纹肌肌节肌动蛋白(αa-SCA)对胚龄9~17d小鼠胚胎心连续切片进行免疫组织化学或免疫荧光显色;免疫印迹检测胚龄11、13、15、10d和17d小鼠胚胎心组织中Cx43的含量变化。结果:胚龄9~10d,仅在左心室腹侧壁及原始小梁最先检测到Cx43弱阳性表达。随着发育,Cx43在心房心室阳性范围逐渐扩展,阳性表达逐渐增强。而在某些特定部位,如窦房结、房室管、房室结和房室束等始终未见Cx43阳性染色。结论:Cx43在胚胎心的时空表达模式与心工作心肌和传导系心肌的发育及兴奋传导功能相适应。Objective: To investigate the spatial and temporal expression patterns of connexin 43 in the mouse embryonic hearts. Methods: Serial transverse sections of mouse embryonic hearts from El) 9 to ED 17 were stained immunohistochemically or immunofluorescently with antibodies against connexin 43 (Cx43), myosin heavy chain (MHC) and α-sarcomeric actin (α-SCA) ; Cx43 protein content was examined by Western blotting in ED 11, 13, 15, 16 and 17 of the embryonic hearts. Results: During ED 9 to 10, the weak positive expression of Cx43 became first detectable only in the ventral wall of the left ventricle and primary trabeculum. With development, Cx43 positve expression in the atrium and ventricle gradually expanded and increased, whereas it remained undetectable in some myocardium, such as the sinoatrial node, atrioventricular canal, atrioventricular node and atrioventricular bundle. Conclusion:The spatial and temporal expression patterns of Cx43 in the mouse embryonic heart correlates with the development of the working myocardium and cardiac conduction system myocardiurn and with the conduction of excitation.
