摘 要:目的:探讨何首乌饮对运动疲劳大鼠睾丸组织黄体生成素受体(LHR)的影响。方法:复制运动疲劳动物模型,测定睾酮水平,采用RT-PCR、免疫印迹和免疫组织化学法检测各组睾丸组织LHR的表达变化。结果:LHR免疫组化阳性颗粒主要表达于问质细胞以及精母细胞胞膜和胞质,何首乌饮预防组和何首乌饮治疗组阳性信号较强,模型组最弱。LHR蛋白和mRNA表达变化为:何首乌饮预防组、何首乌饮治疗组明显高于对照组和何首乌饮组,模型组明显低于对照组。结论:何首乌饮可以提高大鼠睾丸组织LHR的表达。Objective: To explore the effect of Heshouwuyin on the gene expression of luteinizing hormone receptor (LHR) in the testis tissue of rats with exercised-induced fatigue. Methods: Fifty SD rats were divided into 5 groups randomly (n= 10), includinga normal control group, a Heshouwuyin administered group, a model control group, a Heshouwuyin treated group and a Heshouwuyin prevented group. Beckmancoulter Unicel Dxl 800 was used to detect the level of serum testosterone according to the manufacture's instructions. RT PCR, Western blotting and immunohistochemistry were used to observe the differential expression of LHR. Results: LHR protein was expressed in the membrane and cytoplasm of Leydig cell and spermatocyte. Compared with the normal control group, the expression of LHR protein and mRNA in the model control group decreased, and the expression of LHR protein and mRNA in the Heshouwuyin treated and Heshouwuyin prevented groups increased more significantly than that in the normal control group and Heshouwuyin administered groups. Conclusion: The expression of LHR can be up-regulated by Heshouwuyin.
