摘  要:目的:观察APP/PS1双转基因阿尔茨海默病(AD)模型小鼠(APP/PS1dtg)蓝斑去甲肾上腺素能神经元的数量和形态变化。方法:选用雌性APP/PS1dtg和同龄野生型小鼠,分为年轻组(5~6月龄)和老年组(16~17月龄)。取小鼠脑组织冷冻切片,采用酪氨酸羟化酶(TH)免疫组织化学显色显示蓝斑去甲肾上腺素神经元、无偏差体现学方法定量分析TH阳性神经元总数及细胞体积等的变化。结果:免疫组织化学显色显示,与野生型小鼠比较,老年组APP/Ps1dtg蓝斑TH阳性神经元胞体变大,细胞总数量减少23%,并可见形态异常的TH阳性纤维。此外,在APP/PS1dtg,蓝斑TH阳性神经元的平均细胞体积老年组比年轻组增加,并与蓝斑总体积呈正相关。结论:老年组APP/PS1dtg小鼠蓝斑TH阳性神经元和纤维发生退行性改变,这些改变与AD患者蓝斑的病理改变相似。Objective: To investigate quantitative and morphological change of noradrenergic neuron in locus coeruleus of double transgenic APP/PS1 (APP/PS1 dtg) Alzhmeimer's disease (AD) model mice. Methods: Mice were divided into two groups:a young group (5-6 months old APP/PS1 dtg and wild type mice) and an aged group (16-17 months old APP/PS1 dtg and wild type mice). The brain cryosections were cut and stained with tyrosine hydroxylase (TH) immunohistochemistry, and unbiased stereology was used to analyze the total number of TH positive neurons and its morphological changes in the locus coeruleus. Re- sults: Immunohistochemiically, there was a significant 23% reduction in the total number of TH-positive neurons and qualitatively abnormal TH-positive fiber morphology and swollen body of TH-positive cells in the locus coernleus of the old-aged APP/PS1 dtg mice compared with the old-aged wild type mice Furthermore, in the group of APP/PS1 dtg mice, there was a significant age-related increase in mean cell volume of TH-positive neurons, and the mean cell volume of TH-positive neurons in the old-aged APP/PS1 dtg group significantly increased compared with dtg young APP/PS1 dtg type mice. Conclusion: These locus coeruleus-related changes show resemblance to those present in the brain of AD patients at postmortem examination
