摘 要:目的探讨围产期低剂量氯化三丁基锡(tributylin chloride,TBTCl)暴露对子代成年雄性小鼠发育及血清、睾丸中雌激素(E2)和睾酮(T)水平的影响是否具有持续性。方法将20只妊娠6 d的SPF级昆明小鼠采用经口灌胃方式染毒0(对照,玉米油)、1、10、100μg/kg TBTCl溶液,每组5只,每天1次,染毒至哺乳期[出生后21 d(PND21)]结束。仔鼠断乳后每周称量一次体重。至PND 84时,每组随机抽取10只雄性仔鼠,测定脾脏、肝脏、肾脏、胸腺的重量,并计算脏器系数;测定血清和睾丸匀浆组织中E2和T水平。结果与对照组比较,100μg/kg TBTCl组子代成年雄性小鼠在PND0、49、56、63、70、77时的体重均显著降低(P<0.05,P<0.01),PND84时各剂量TBTCl染毒组体重均无显著改变;脾脏重量及其脏器系数均显著升高(P<0.05),而肾脏重量及其脏器系数均显著降低(P<0.05),而肝脏和胸腺重量及其脏器系数均无显著变化;100μg/kg TBTCl染毒组睾丸组织匀浆中E2水平显著降低(P<0.05),而血清中E2和T水平及睾丸组织匀浆中T水平均无显著变化。结论围产期低剂量TBTCl暴露对子代成年雄性小鼠的发育和睾丸组织中的E2水平具有持续的毒性作用。Objective To explore the continuity of the effects of perinatal exposure to low-close tributylin chloride (TBTC1) on development and the levels of estrogen (E2) and androgen (T) in serum and intratesticular of the adult male offspring mice. Methods The SPF adult pregnant Kunming mice were randomly divided into four groups, five in each group, and were given TBTC1 (100,10 and 1 μg/kg body weight,respectively, once per day) by gavage from the 6th day of gestation to the end of lactation(PND21), the mice in control were received an equal volume of corn oil as vehicle(5 ml/kg). The body weights of the male mice after ablactation were weighed every week. On PND84,10 male offspring mice were randomly selected in each group. The liver, spleen, kidney and the thymus were separated and weighed for account of viscera coefficient, the levels of estrogen(E2) and androgen(T) in serum and intratesticular were measured respectively. Results Compared with the control group, the body weight of the 100μg/kg group significantly decreased on PND0, PND49, PND56, PND63, PND70 and PND77 (P〈0.05, P〈0.01 ). The weight and the viscera coefficient of the spleen revealed significant increase, while the kidney revealed significant decrease in 100 μg/kg group (P〈0.05). There were no significant differences in the levels of estrogen(E2) and androgen(T) in serum and intratestieular(P〉0.05). However, the levels of estrogen(E2) and androgen(T) in serum and intratesticular in 100 μg/kg group decreased significantly compared with the control group (P〈0.05). Conclusion Perinatal exposure to low-dose TBTC1 may affect the male offspring development and the level of sex hormone in testis abidingly in mice.
