摘 要:目的观察健脾补肾方对Lewis肺癌小鼠肿瘤细胞的抑制作用及对Ki-67表达影响。方法采用Lewis肺癌移植瘤C57BL/6小鼠模型观察健脾补肾方抗肿瘤的作用,并采用HE染色观察各组肿瘤细胞的凋亡情况,免疫组化法检测肿瘤组织中Ki-67增殖抗原蛋白的表达情况。结果与对照组相比,健脾补肾方低剂量组能增加荷瘤小鼠胸腺指数,有统计学意义(P<0.05)。低、高剂量健脾补肾方与CTX合用,能对CTX所致荷瘤小鼠胸腺萎缩有一定的拮抗作用。健脾补肾方能下调Lewis肺癌小鼠肿瘤组织Ki-67蛋白的表达水平。结论健脾补肾方对小鼠Lewis肺癌具有显著的抗肿瘤作用,以健脾补肾方高剂量加环磷酰胺组最佳。其作用机制可能与该药能降低肿瘤组织Ki-67蛋白的表达水平有关。Objective To investigate the anti-tumor effect and mechanism of Jian-pi-bu-shen-fang.Methods The mouse model of C57BL /6 bearing lewis lung carcinoma was established to investigate the anti-tumor effect of Jian-pi-bu-shen fang.The expressions of Ki-67 protein were detected in tumor tissues by three-step immuno-histochemistry staining.Results The low dose of Jian-pi-bu-shen-fang group could improve the thymus index of the mice bearing lewis lung carcinoma,and had significance in statistics with the model group(P 〈 0.05).Jian-pi-bu-shen-fang down-regulated the expression of Ki-67 protein in carcinoma tissues from the mice bearing lewis lung carcinoma.Conclusion Jian-pi-bu-shen-fang has an anti-tumor effect,and the effects of the the high dose of Jian-pi-bu-shen-fang and CTX group are optimum.The molecular mechanism of anti-tumor activity can concerned with down-regulation of the Ki-67 in tumor tissues.
