补中益气汤对创伤大鼠应激反应调理作用实验研究

摘　　要：目的:研究补中益气汤对创伤大鼠应激反应的影响,初步探索其作用机制。方法:将30只SD大鼠随机分成3组,每组均为10只,即分成Ⅰ组正常对照组、Ⅱ组模型对照组、Ⅲ组中药治疗组。模型对照组和中药治疗组制成骨折创伤模型,伤后1h开始给Ⅰ组、Ⅱ组经胃灌注生理盐水,给Ⅲ组经胃灌注中药,每次均为0.5mL,每日2次。于创伤后6h、1、3、5、7天采静脉血测定应激激素COR、NE和细胞因子TNF-α、IL-4的水平。结果:Ⅱ组COR在各时间点较Ⅰ组显著升高(P<0.05或P<0.01),上升缓慢而持久;Ⅱ组NE在伤后6h、1天较Ⅰ组显著升高(P<0.01),整个时间点NE上升快,但下降也快;Ⅱ组TNF-α在伤后显著升高(P<0.01),在整个时间点上无显著变化;Ⅱ组细胞因子IL-4在创伤后明显降低(P<0.01),且回升缓慢。Ⅲ组对血中应激激素COR在伤后3、5、7天的升高有显著控制作用,而NE在伤后6h、1天的升高有显著控制作用,与Ⅱ组比较差异有显著性(P<0.05或P<0.01)。Ⅲ组对血中细胞因子TNF-α的升高和对细胞因子IL-4的降低均有显著控制作用,与Ⅱ组比较差异均有显著性(P<0.05或P<0.01)。结论:补中益气汤能有效调理创伤后应激反应的发生和发展。Objective ： To study effect of Buzhong Yiqi Decoction on stress response in traumatic rats,preliminary exploration of its mechanism of action. Methods ： 30 SD rats were randomly divided into 3 groups, each group had 10 only, which is divided into the normal control group （ I group ）, model control group （ 1I group ）, treated with Chinese medicine （m group ）. 11 group and m group made fracture trauma model, and lh after injury began to I group and II group by gastric perfusion with saline, to III group by gastric perfusion in traditional Chinese medicine, each time 0.SmL, 2 times a day. After trauma, 6h 1,3, 5,7d collecting venous blood determination stress hormones of COR, NE and cytokines of TNF-oL , IL-4 levels. Results ： In ]I group COR at each time point compared with I group increased significantly （ P〈0.05 or P〈0.01 ）, rises slowly and lasting; in II group NE after injury in 6h, ld compared with I group increased significantly （ P〈0.01 ）, the whole point of time NE rises quickly, but decreased quickly; in II group TNF-a after trauma significantly elevated （P〈0.01）, the point in time had no significant change; cytokine IL-4 in posttraumatic decreased significantly （P〈0.01）, and a slow pick up. III Group on blood stress hormones COR after injury in 3,5,7d had significant control effect of elevated, and NE after injury in 6h, ld increased significantly control function, compared with II group there was a significant difference （ P〈0.05 or P〈0.01 ）. Group III on blood cytokine TNF - a raised and on cytokine of IL-4 reduces significantly control function, compared with group had significant difference （ P〈0.05 or P〈0.01 ） .Conclusion ： Buzhong Yiqi Decoction can effectively adjust posttraumatic stress reactions occur and development.
