摘  要:目的:探讨小鼠孕期酒精暴露对仔鼠齿状回苔藓细胞的影响,并进一步观察神经鞘磷脂合成酶2基因敲除(sphingomyelin synthase 2 knockout,SMS2-/-)与孕期酒精暴露和苔藓细胞丢失之间的关系。方法:采用PCR法对各分组仔鼠进行鉴定,利用SMS2-/-小鼠建立孕期酒精暴露模型,收集P14仔鼠,进行水迷宫实验,观察仔鼠寻台时间;然后利用免疫荧光染色法观察各分组仔鼠齿状回苔藓细胞数量的变化,免疫印迹技术检测P14仔鼠海马组织GluR2/3蛋白的相对表达量。结果:(1)与对照组相比,酒精组仔鼠的寻台时间相对较长(P<0.05),而SMS2-/-组仔鼠的寻台时间与野生组无明显的差异(P>0.05);(2)酒精组仔鼠齿状回GluR2/3的阳性细胞数明显少于对照组(P<0.05),且SMS2基因敲除后,与野生组仔鼠相比,SMS2-/-组仔鼠齿状回GluR2/3的阳性细胞数无明显差异(P>0.05)。结论:(1)酒精可诱导齿状回苔藓细胞的丢失,且降低GluR2/3的阳性表达;(2)SMS2基因的敲除对酒精诱导苔藓细胞丢失的作用相对较小;(3)苔藓细胞的丢失在一定程度上降低近期记忆功能,酒精可促进记忆功能的恶化。Objective: In order to investigate the effects of alcohol exposure during pregnancy of mouse on mossy cells of dentate gyrus in offspring, and further observe the relationship of the sphingomyelin synthase 2 gene knockout ( SMS2^-/^- ) with the alcohol exposure during pregnancy and loss of mossy cells. Methods : Using SMS2^-/^- mice to establish the model of prenatal alcohol exposure. Collection of P14 rats, the Water Maze test was used to observe that the offspring find plat- form time. In the meantime, the loss of mossy cells in dentate gyms was investigated before and after alcohol exposure with immunofluorescent labeling. The expression of GluR2/3 in hippocampus was also tested with Western blot analysis. Results : ( 1 ) Compared with the control group, the rats of alcohol group found platform time was relatively long ( P 〈 0.05 ), and it was no significant difference between SMS2^-/^- group mice and Wild-type group ( P 〉 0.05 ). ( 2 ) The GIuR2/3 positive cells in dentate gyms of alcohol group offspring were significantly less than those of control group ( P 〈0.0.5 ) , and after SMS2 knorkout, compared with the wild-lype group, the GIuR2/3 positive veils of SMS2^-/^- group were no significant difference (P 〉 0.05 ). Conclusion: ( I ) Alcohol can induee the loss of the mossy cell in dentate gyrus, and reduce the positive expression of the GIuR2/3 ; (2) SMS2 gene knockout had a relatively small role to mossy cells lost; (3) To a certain extent, loss of mossy tells reduee the rerent memory function, and alcohol can promote the deterioralion of memory function.
