补中益气汤对小鼠肠道mBD-2表达的影响

摘　　要：目的研究补中益气汤对小鼠肠道上皮细胞天然免疫效应分子β防御素-2(mouseβ-defensin 2,mBD-2)表达的影响。方法取SPF级C57BL/6小鼠,随机分为正常对照组和补中益气汤高、中、低剂量组,给予生理盐水和不同剂量补中益气汤(50,25,12.5 g.kg-1)灌胃,7 d后取小鼠肠道组织,分别用RT-PCR法和免疫组化法检测小鼠肠道上皮细胞mBD-2 mRNA和蛋白表达。结果正常对照组小鼠肠道组织不表达mBD-2。经补中益气汤刺激后,在mRNA水平和蛋白水平均检测到肠道上皮细胞mBD-2的表达,补中益气汤高剂量组作用效果明显,与中、低剂量组比较差异有统计学意义(P<0.05,P<0.01)。结论补中益气汤在体内可以诱导小鼠肠道上皮细胞天然免疫效应分子mBD-2的表达,这可能是其抗炎抑菌的作用机理之一。Objective To investigate the effects of Buzhong Yiqi Decoction on the expression of beta-defensin 2 （mBD-2）, an effector molecule of innate immune, in mouse intestinal epithelial cells in vivo. Methods C57BL/6 mice were randomly divided into normal control group and three experiment groups. The mice of experiment groups were administered with different doses of Buzhong Yiqi Decoction（50, 25 and 12.5 g.kg-1） orally. RT-PCR and immunohistoehemistry were used to detect mBD-2 mRNA and protein expression in mouse intestinal epithelial cells. Results mBD-2 expression was not detected in the normal control group. However, after treatment with Buzhong Yiqi Decoction, mBD-2 mRNA and protein expression was detectable, and the expression levels were higher in high-dose group than those in moderate- and low-dose groups （P 〈 0.05 or P 〈 0.01 ）. Conclusion mBD-2 expression is induced by Buzhong Yiqi Decoction in intestinal epithelial cells in vivo, which may be one of the anti-inflammation and antibacterial mechanisms of Buzhong Yiqi Decoction.
