摘 要:目的观察明睛颗粒对激光诱发脉络膜新生血管(CNV)后骨髓细胞动员的影响,探讨明睛颗粒防治脉络膜新生血管的作用机制。方法将氪激光诱导脉络膜新生血管的C57BL/6小鼠随机分为治疗组和模型组,以正常小鼠作正常对照组。采用流式细胞仪检测小鼠在治疗后7,14,28 d外周血的CD34+细胞百分数,酶联免疫吸附法检测外周血粒-巨噬细胞集落刺激因子(GM-CSF)、血管生长因子(VEGF)的含量,并在治疗后28 d用眼底荧光血管造影观察脉络膜新生血管情况。结果治疗后7,14 d,模型组外周血CD34+细胞数明显高于治疗组及正常对照组(P<0.05);治疗组外周血GM-CSF及VEGF含量均明显低于模型组(P<0.05);治疗后28 d,治疗组CNV渗漏面积明显小于模型组(P<0.05)。结论初步显示明睛颗粒能在早期抑制CNV小鼠外周血骨髓干细胞的动员,提示其对CNV的防治作用可能与抑制骨髓来源细胞参与CNV形成有关。Objective To observe the effect of Minging Granula on the mobilization of bone-marrow derived cells after laser-induced choroidal neovascularization (CNV), and to investigate the possible preventive mechanism of Mingjing granula for laser-induced CNV. Methods C57BL/6 mice with CNV induced by krypton laser were randomly divided into the model group and treatment group. Ten normal mice were used as the normal control without any medication. The amount of CD34+ cells in peripheral blood were detected by flow cytometer on day 7, 14 and 28 after treatment. The levels of granulocyte-macrophage colony-stimulating factor(GM-CSF) and vascular endothelial growth factor (VEGF) were determined by enzyme-linked immunosorbent assay(ELISA) on day 7, 14 and 28 after treatment, and the CNV leakage area was examined with fluorescein fundus angiography(FFA) on day 28 of the treatment. Results The amount of CD34+ cells in the model group were significantly higher than that of the treatment group and normal group on day 7 and 14(P 〈 0.05), and there was no significant difference between the treatment group and the normal group(P 〉 0.05 ). The levels of GM-CSF and VEGF in the treatment group were significantly lower than those of the model group and normal group on day 7 and 14(P 〈 0.05). The CNV leakage area of FFA in the treatment group was significantly smaller than that of the model group on day 28 after treatment(P 〈 0.05 ). Conclusion It is initially indicated that Mingjing Granula could inhibit the mobilization of bone-marrow derived cells in laser-induced CNV mice at the early stage, which may be the possible preventive mechanism of Mingjing Granula for laser-induced CNV.
