摘  要:目的探讨补肾活血方、补肾方、活血方对血管性痴呆小鼠学习记忆能力及脑组织碱性成纤维细胞生长因子(bFGF)mRNA表达的影响。方法采用脑缺血再灌注法复制血管性痴呆小鼠模型,以尼莫地平为阳性对照组,观察补肾方、活血方、补肾活血方3组小鼠水迷宫法行为学、脑组织bFGF含量及bFGF mRNA表达的变化。结果与假手术组比较,模型组小鼠学习与记忆能力下降,表现为游全程时间延长,错误次数增加(P<0.01),脑组织bFGF含量和bFGF mRNA表达相对含量明显升高(P<0.05或P<0.01);与模型组比较,各治疗组学习成绩和记忆成绩均有明显提高(P<0.05或P<0.01),脑组织bFGF含量和bFGF mRNA表达相对含量明显升高(P<0.01);各治疗组比较,补肾活血组的学习成绩与记忆成绩优于其他治疗组(P<0.05),bFGF含量及脑组织bFGF mRNA表达相对含量均高于活血组(P<0.05),与补肾组比较无统计学差异(P>0.05)。结论补肾方、活血方、补肾活血方均可通过促进脑组织bFGF mRNA表达,提高脑组织bFGF含量,改善血管性痴呆小鼠学习记忆能力,且补肾活血方在促进小鼠学习记忆能力方面显著优于补肾方和活血方,在调节bFGF含量及bFGFmRNA表达方面明显优于活血方。Objective To probe the effect of Bushen Recipe, Huoxue Recipe, and Bushen Huoxue Recipe on abilities of learning and memory as well as the expression of basic fibroblast growth factor(bFGF) mRNA in brain issue of vascular dementia mice. Methods We established the mice model of vascular dementia through the operation of cerebral ischemia reperfusion, observed the praxiological changes by the means of water maze, and examined the content of bFGF and the expression of bFGF mRNA in brain tissue in mice after treated with Bushen Recipe, Huoxue Recipe, and Bushen Huoxue Recipe, respectively. Results Compared with the sham operation group, the ability of learning and memory reduced in the model group, which was characterized with the prolongation of time for swimming the whole process and the increase of times of error (P 〈 0.01 ), and the content of bFCF and the expression of bFGF mRNA in brain issue was increased (P 〈 0.05 or P 〈 0.01 ). Compared with model group, the learning and memorizing performance scores were improved (P 〈 0.05 or P 〈 0.01), and the content of bFGF and the expression of bFGF mRNA were increased (P 〈 0.01) in brain tissue of each treatment group. The learning and memorizing performance scores of Bushen Huoxue Recipe group were superior to those in the other two treatment groups (P 〈 0.05), and the contents of bFGF and the expression of bFGF mRNA in brain issue were superior to that in Huoxue Recipe(P 〈 0.05 ) but had no different significant with Bushen Recipe group (P 〉 0.05). Conclusion Bushen Recipe, Huoxue Recipe, and Bushen Huoxue Recipe all could increase the content of bFGF and improve the learning and memory abilities through promoting the expression of bFGF mRNA. The improvement of the learning and memory by Bushen Huoxue Recipe is superior to that of Bushen Recipe and Huoxue Recipe, and its regulation of bFGF content and bFGF mRNA expression is superior to that of Huoxue Recipe.
