摘  要:目的优选聚酰胺纯化枳实总黄酮的工艺条件。方法采用紫外分光光度法,以枳实总黄酮的吸附率、解吸率作为考核指标,筛选出聚酰胺纯化枳实总黄酮的最佳工艺参数。结果最佳纯化工艺条件:上样液浓度为3.0 mg.mL-1,pH值为6.0~7.0,上样量为1 BV,吸附1.5 h后,先用2 BV的水洗脱,再用3 BV的80%乙醇洗脱。结论聚酰胺树脂对枳实总黄酮的纯化效果好,成本低,可工业化推广。Objective To obtain the optimal technique data for purification of the total flavonoids in Fructus Aurantii Immaturus by polyamide resin. Methods Ultraviolet spectrometry method was adopted, and the absorption quantity of the total flavonoids and the yield of desorption materials with ethanol were determined to optimize the process conditions of polyamide resin in purification of total flavonoids. Results The optimal adsorption and desorption conditions were as follows: the total flavonoids concentration was 3.0 mg.mL-1, the sample volume was 1BV, and pH value was 6.0 to 7.0. The sample solution was rested for 1.5 hour in the chromatographic column of polyamide resin, eluted with 2BV of deionized water, and then eluted again with 3BV of 80 % alcohol. Conclusion Polyamide resin has better purifying effect on the total flavonoids in Fructus Aurantii Immaturus, which is suitable for large scale industrial production.
