摘  要:以大花萱草‘莎蔓’子房为外植体,对其组织培养中愈伤组织诱导分化、试管苗繁殖的激素配比对繁殖效率的影响以及生根培养基的设置和驯化栽培方法等关键技术参数进行系统研究。结果表明:‘莎蔓’子房愈伤组织诱导最佳培养基为MS+4mg/L 6-BA+0.3mg/L IBA+1mg/L 2.4-D,不定芽分化率可达49.2%;在3mg/L 6-BA、(0.3~0.5)mg/L IBA时,繁殖系数可达5.8~6.4倍。适宜的生根培养基为1/2MS+0.4mg/L NAA+20g/L蔗糖+6.5g/L琼脂,平均试管苗生根数达5.7条。组培过渡苗栽培基质为粗河沙、蛭石+泥炭土、泥炭土均可,条件控制在温度24~28℃,相对湿度85%~90%,移栽成活率可达到90%。Using ovary of Hemerocallis hybrida ‘Shaman’as explant,callus induction and differentiation,the effect of hormone combinations on reproductive efficiency of plantlets,setting of rooting medium,the key technical parameters of domestication and cultivation in the process of its tissue culture were systematically studied.The results showed that the best culture medium of ‘Shaman’callus differentiation was MS+4mg/L 6-BA+0.3mg/L IBA+1mg/L 2.4-D and adventitious bud differentiation rate reached 49.2%.When 6-BA concentration was 3mg/L and(0.3~0.5) mg/L IBA,propagation coefficient would be possible to reach 5.8~6.4times.The suitable rooting medium included 1/2MS+0.4 mg/L NAA+20g/L sucrose+6.5g/L agar,the average rooting number reached 5.7roots.The substrate for transitional cultivation of tissue culture seedling was thick riversand,vermiculite+peat soil,peat soil,when the temperature was 24~28℃,the relative humidity was 85%~90%,survival rate of tube seeding reached 90%.
