摘 要:利用原子荧光分光光度法测定了桑葚果酒中硒的含量。采用单因素考察法优化桑葚果酒中芦丁、槲皮素的提取条件,采用HPLC法测定芦丁、槲皮素的含量,色谱柱为Kromasil C18(4.6 mm×150 mm,5μm),测定芦丁以乙腈-0.1%磷酸(体积比19∶81)为流动相,测定槲皮素以甲醇-0.4%磷酸(体积比1∶1)为流动相,流速为1.0 mL/min,检测波长为360 nm,柱温为30℃。结果表明,所选桑葚酿造的果酒中硒含量为(0.015 3±0.001 2)mg/mL。槲皮素在2.02~40.40μg呈良好线性关系(R2=0.999 9),平均回收率为98.88%,RSD为1.19%(n=5)。试验结果为安康富硒桑葚果酒的开发提供一定的理论依据。Atomic fluorescence spectrum was used to determine content of selenium of mulberry wine.Single-factor analysis was applied to optimize the extract condition of rutin and quercetin in selenium-rich mulberry wine,and HPLC method was used in the determination of rutin on C18 column(4.6 mm×150 mm,5 μm)with a mixture of acetonitrile-0.1% phosphoric acid(19∶81,V/V) and a mixture of methanol-0.4% phosphoric acid(1∶1,V/V) as a mobile phase at the flow rate of 1.0 mL/min.Rutin and quercetin were detected at 360 nm,and column temperature was 30 ℃.The results showed that the selenium content of mulberry wine was(0.015 3±0.001 2) mg/mL.The linearity of quercetin was in the range of 2.02~40.40 μg with R2=0.999 9.The average recovery of quercetin was 98.88% and RSD was 1.19%(n=5).This experiment provided certain experiment basis for development of Se-rich mulberry wine.
