摘  要:建立了比色法测定刺槐(Robinia pseudoacacia L.)花提取物总皂苷含量的方法。以人参皂苷Rg1作为对照品,通过正交设计对香草醛-高氯酸比色法的试验条件进行优化,确定最佳比色条件为:60℃水浴挥干溶剂后,加入0.2 mL 7.0%香草醛-冰乙酸液和1 mL高氯酸,于60℃水浴20 min,测定波长为545nm。样品在0~0.24 mg范围内呈良好的线性关系,回归方程为y=3.547 0x+0.010 9(R2=0.998 9),平均加样回收率为100.07%(RSD=2.10%,n=7),测得刺槐花提取物中总皂苷平均含量为2.177%。该方法简便、准确、重现性与稳定性较好,可用于测定刺槐花提取物总皂苷含量。Study was carried out to establish the method of determining saponins content in Robinia pseudoacacia L.flowers(RPF) by colorimetry.With Ginsenoside Rg1 as standard material,the condition of vanillin-perchloric acid colorimetry was optimized by orthogonal design.The suitable conditions for colorimetry were obtained as follows : the sample solution was evaporated by water bath heating at 60 ℃,and was mixed with 0.2 mL acetic acid containing 7.0% vanillin and 1 mL perchloric acid,and reaction mixture was incubated at 60 ℃ for 20 min.Detection wavelength was 545 nm.A good linear relationship was achieved in the range of 0~0.24 mg with the regression equation of y=3.547 0x+0.010 9(R2=0.998 9),and the average recovery was 100.07%(RSD=2.10%,n=7).The average content of total saponins in RPF was 2.177%.This method was simple,accurate,excellently stable and repeatable,and was suitable for the determination of total saponins in RPF.
