摘  要:目的改进并验证金黄色葡萄球菌细胞膜20肽诱导的IgAN动物模型,同时通过对模型中氧化应激标志物的检测分析,进一步探讨氧化应激在IgA肾病发病机制中的可能作用。方法①隔周用含有20肽的麦芽糖结合蛋白与弗氏不完全佐剂混合液进行皮下免疫,第9周起每周取尿测定,21周处死动物后肾组织进行光镜,免疫荧光,电镜检测。②取动物肾组织制备组织匀浆,分光光度法检测其中SOD、CAT、MDA、GSH、GSH-Px含量。结果①模型鼠的尿和肾组织的检测结果与人IgA肾病极为相近。②模型组CAT,GSH-Px含量低于其它对照组,具有统计学意义(P<0.05),模型组MDA含量高于其它对照组,且具有统计学意义(P<0.05)。结论金葡菌诱导的此IgA肾病模型,与人IgA肾病具有高度相似性,且具有可重复性。氧化应激反应参与了IgA肾病动物模型的发病机制。Objective To improve and confirm the IgA nephropathy animal model which was induced by Staphylococcus aureus cytomembrane 20 peptide, and to discuss the possible role of oxidative stress in the pathogenesis of IgAN. Methods ①Balb/c mice were immunized sub- cutaneouly with 20 peptide-MBP-fusion protein and Freund' s incomplete adjuvant every two weeks. The urines were collected and detected every week from the ninth week. Mice were sac- rificed at the end of 21weeks and sections of the major organs were processed. Histological ex- amination were done by light microscopy and immunofluorescence and electron microscopy de- tection. ②Miee homogenate was prepared for detecting the different concentration of SOD, CAT, MDA,GSH and GSH-Px with spectrophotometric method. Results ③The urinalysis and nephridial tissue histopathological features in IgAN animal models were extremely similar to hu- man IgAN. ④The concentration of CAT, GSH-Px and MDA between model group and control groups had statistical significance ( P 〈 0. 05 ). Conclusion The IgAN model induced by Staphylococcus aureus cellular membrane antigen 20 peptide has a repeatability, and has signifi- cant similarity with human IgAN. Oxidative stress marker may participate the pathogenesis of IgA nephropathy model.
