摘 要:目的通过血液自体荧光光谱分析探讨黄芪多糖对H22荷瘤小鼠肿瘤抑制作用机制。方法将H22荷瘤小鼠随机分为肿瘤组、5-氟尿嘧啶(5-Fu)组和黄芪多糖(APS)组,采用970CRT荧光分光光度计,以312 nm和405 nm波长的光激发,检测其血清自体荧光光谱和红细胞中卟啉自体荧光光谱,分别测量波峰的位置、形态、荧光强度并分析。结果血清自体荧光实验中3组均在380 nm和460 nm出现峰值,5-Fu和APS组相对荧光强度强于肿瘤组(P﹤0.05);红细胞卟啉自体荧光实验中,肿瘤组的锌卟啉峰值与原卟啉峰值比大于1,而两用药组均小于1(P﹤0.05)。结论 APS可通过调节能量代谢和一些特定蛋白质的含量产生抑癌作用。Objective (APS) on H22 tumor- To study mechanism of tumor inhibition of astragalus polysaccharide bearing mice by blood auto-fluorescence. Methods The H22 tumor- bearing mice were randomly divided into the tumor group, the 5-Fu group, and the astragalus polysaccharides (APS) group. Serum auto-fluorescence spectrum and porphyrin auto-fluores- cence spectrum in red cells of I-I22 mice in different groupswith excitation wave length of 312 nm and 405 nm were measured by use of fluorescence spectrophotometer, and the peak posi- tion, shape, fluorescence intensity in three groups were measared and analysed. Results In the serum auto-fluorescence experiment, three groups had peaks in both 380 nm and 460 nm, relative fluorescence intensity in drug groups was stronger than that in the tumor group (P 〈 0. 05) ; in the porphyrin auto-fluorescence spectrum of red ceils experiment, the ratio of the peak of Zn-porphyrin with that of protoporphyrin(PPIX) was more than 1 in the tumor group, but in the other two groups were less than 1 (P 〈 0. 05 ). Conclusion APS is generated the role of anti-tumor by regulating energy metabolism and some specific content of protein.
