摘 要:核转录因子κB(NF-κB)在调节细胞免疫功能及细胞增殖和存活方面均发挥着重要的作用。本研究采用高致病性猪繁殖与呼吸综合症病毒(HP-PRRSV)GD08-1株和经典株PRRSV GD-XH分别感染Marc-145细胞,于感染后不同时间提取胞浆蛋白和核蛋白,通过电泳迁移率(EMSA)检测NF-κB与DNA结合活性及western blot检测胞浆蛋白中NF-κB及其抑制因子(IκB)的表达情况。HP-PRRSV株和PRRSV经典株感染Marc-145核蛋白的EMSA结果中均出现NF-κB与DNA探针的结合条带;western blot结果表明,HP-PRRSV株和PRRSV经典株感染Marc-145细胞并未引起NF-κB表达量的变化,但两者均引起了IκB表达水平的下降,由此推测,HP-PRRSV株和经典PRRSV株感染均能够刺激Marc-145细胞NF-κB活化入核内,PRRSV感染Marc-145细胞引起的NF-κB的活化是通过IκB的降解来实现的。Nuclear factor-kappaB (NF-κB) is a critical regulator of immune function as well as cell proliferation and survival. In this study, Marc-145 cells were infected with highly pathogenic pocine reproductive and respiratory syndrome virus (HP-PRRSV) GD08-1 strain and PRRSV GD-XH strain. The cytoplasmic and nuclear protein were extracted from Marc-145 cells at different post infection, respectively. DNA binding activity of NF-κB was assessed by EMSA and the protein expressing levels of NF-κB and IKBα in cytoplasmic from Marc-145 cells was assessed by western blot. The results indicated that both NF-κB in nuclear proteins from HP-PRRSV and PRRSV infected Marc-145 cells were positively binding to DNA. However, the cytoplasmic protein levels of both NF-κB had no changed in virus infected Marc-145 cells, but IKBα protein levels were degraded in both viruses infected cells. These results demonstrated that both HP-PRRSV and PRRSV infection resulted in an increase in NF-κB nuclear translocation and NF-κB-DNA binding activity mediated by IKBα degradation in MARC-145 cells.
