摘  要:为研究甘草酸苷(GL)对小鼠单核巨噬细胞细胞因子分泌的影响,本实验以100μg/mL的GL溶液处理小鼠单核巨噬细胞(RAW264.7),培养后收集培养上清,采用ELISA法测定其水平。结果显示,GL能够显著提高培养上清中IFN-α(p<0.01)、IFN-β(p<0.05)和IFN-γ(p<0.01)水平,降低促炎因子IL-1β(p>0.05)、IL-6(p<0.05)、IL-12(p>0.05)和CXCL10(p<0.05)含量,并增加TGF-β的分泌(p>0.05)。这表明,GL可以通过诱导小鼠单核巨噬细胞产生IFN-α、IFN-β和IFN-γ而发挥抗病毒作用,同时能够抑制促炎因子IL-1β、IL-6、IL-12和CXCL10的产生,并促进抑炎因子TGF-β的分泌,从而增强细胞的抗炎能力。本研究为进一步明确GL免疫调控作用机制提供了实验依据。To investigate the effect of glycyrrhizin (GL) on cytokine secretion of murine monocyte macrophage, RAW264.7 cells were cultured in vitro with GL (100 μg/mL) for 20 hours. The culture supernatants were collected and analyzed by detecting the levels of cytokines, including interferon (1FN)-α, IFN-β, IFN-% interleukin (IL)-1β, IL-6, IL-10, IL-12, tumor necrosis factor-α (TNF-α), transforming growth factor-J3 (TGF-β) and CXCL10. The results showed that the GL treatment significantly increased the up-regulated of IFN-α (p〈0.01), IFN-β (p〈0.05) and IFN-γ (p〈0.01), and down-regulated the levels of pro-inflammatory cytokines IL-1β(p〉0.05), IL-6 (p〈0.05), IL-12 (p〉0.05) and CXCL10 (p〈0.05) while increased that of anti-inflammatory cytokine TGF-β (p〉0.05). The findings manifested that GL might exert its antiviral effect in murine monocyte macrophages by inducing IFN-α, IFN-β and IFN-γ production, and enhance anti-inflammatory ability of the cells through suppressing the production of pro-inflammatory cytokines (IL-1β, IL-6, IL-12 and CXCL10) and stimulating the secretion of anti-inflammatory TGF-β. This study also provided experimental evidences for further clarifying the immune-regulating mechanism of GL.
