摘  要:[目的]通过试验来提取地鳖虫多糖,用于中药多糖的开发研究。[方法]采用热水浸提法对地鳖虫多糖进行提取试验,采用单因素试验来选择浸提条件,以浸提时间、料液比、浸提温度、浸提次数这4个因素做正交试验,用苯酚-硫酸进行多糖含量测定。[结果]浸提温度对地鳖虫多糖含量影响最大,其次为浸提次数,浸提时间和料液比影响较小。地鳖虫多糖最佳的提取工艺条件为提取时间1.5h,料液比1∶10 g/ml,浸提温度80℃,浸提次数3次。[结论]影响昆虫水溶性多糖提取率的因素主要有浸提温度、浸提时间、浸提次数和料液比等。随着提取时间的延长,细胞破碎越完全,多糖溶出的越多,所得多糖含量则越高。[ Objective ] To extract Eupolyphaga sinensis polysaccharide through experiment for the research and development of polysaccharides in traditional Chinese medicine. [ Method] The Eupolyphaga sinensis polysaccharides were extracted by hot water, and the extraction conditions were determined by single factor test, an orthogonal experiment was carried out with the four factors of extraction time, solid-liquid ratio, ex- traction temperature and extraction times, the polysaccharides content was determined using phenol-sulfuric acid. [ Result ] Extraction tempera- ture had the largest impact on the extraction of Eupolyphaga sinensis polysaccharides, then followed by extraction time, extraction time and sol- id-liquid ratio. The optimal extraction conditions were 1.5 h of extraction time, 1:10 (g/ml) of solid-liquid ratio, 80 ℃ of extraction tempera- ture, and three times of extraction. [ Conclusion] The main factors influencing the extraction of insect water soluble polysaccharides included extraction temperature, extraction time, extraction times and solid-liquid ratio. With the prolonging extraction time, the cells disrupted com- pletely, more polysaccharide was dissolved out, and the polysaccharides content improved.
