心痛方对心肌缺血模型大鼠MPO活性及PS表达的干预作用

摘　　要：目的探讨心痛方对心肌缺血大鼠模型心肌梗死面积及髓过氧化物酶(MPO)、P选择素(PS)的干预作用。方法 80只清洁级SD大鼠,随机分成假手术组、模型组、心痛方组、欣康组,采用结扎法造成心肌缺血模型,NBT染色法测心肌梗死面积,比色法检测MPO活性,免疫组化法检测PS的表达。结果 1.模型组、心痛方组、欣康组心肌梗死面积、MPO活性及PS表达均显著高于假手术组,差异有统计学意义(P<0.01);2.心肌梗死面积比较,心痛方组、欣康组分别小于模型组(P<0.05),差异均有统计学意义;3.MPO活性比较,心痛方组、欣康组分别低于模型组(P<0.05),而心痛方组低于欣康组(P<0.05),差异均有统计学意义;4.PS表达比较,心痛方组、欣康组分别低于模型组(P<0.05),差异均有统计学意义。结论心痛方能缩小心肌梗死面积,抑制缺血心肌MPO活性及PS表达。在抑制MPO活性方面心痛方优于欣康。Objective To discuss the intervene effects of Xintong decoction on myocardial in- farction areas and MPO, P-selectin （PS） expression levels in myocardial ischemia rats. Methods 80 male SD rats were randomly divided into 4 groups as follows： Sham -operat- ed group,model group,Xintong decoction group and Xinkang group.The rat model of myocardial is- chemia was established by the left desending anterior branch of coronary ligation. Myocardial in- farction area was measured used NBT staining method. MPO activity in ischemic myocardium was detected use colorimetric assay and adhesion molecules PS expression level was observed by immunohistochemical method. Results Compared with Sham-operated group, infarction areas, PS expression levels,MPO activities in model group, Xintong decoction group and Xinkang group were markedly increased （all P〈0.01）. The infarction areas of Xintong decoction group and Xinkang group were significantly lower than those of model group （P〈0.05）. Compared with the model group,MPO activities and PS expression levels of Xintong decoction group and Xinkang group were decreased markedly （P〈0.05）. The MPO activities and PS expression levels of Xintong decoction group were lower than those of Xinkang group （all P〈O.05）. Conclusion Xin- tong decoction can decrease the myocardial infarction area,inhibit the PS expression and MPO ac- tivity,and the inhibition effect on MPO activity of Xintong decoction is superior to that of Xinkang medicine.
