摘  要:目的 探讨肝x受体(LXR)激动剂对脂肪来源的间充质干细胞移植治疗小鼠急性心肌梗死(MI)的影响。方法酶消化法分离出稳定表达报告基因萤火虫荧光素酶(firefly lueiferase,Fluc+)小鼠的脂肪间充质干细胞(AD-MSC^Fluc),流式细胞术检测CD90、CD44、CD34、CD45等细胞表面标志物。雄性近交系小鼠(无报告基因表达)随机分成以下4组(每组10只):(1)假手术组;(2)MI+PBS组;(3)MI+AD-MSC^Fluc+组;(4)MI+AD-MSC^Fluc+LXR激动剂(T0901317)干预组,每组分别进行相应的干预。生物发光成像(BLI)检测移植后AD.MSC的存活情况,小动物超声检查评价小鼠心脏功能。结果流式细胞术结果显示脂肪间充质干细胞CD44、CD90阳性,CD34、CIM5阴性。光学成像结果显示AD-MSC^Fluc的光学信号强度与细胞数量呈正相关(r^2=0.98)。术后在体生物发光成像结果显示术后第7、14、2l、28天MI+AD-MSC^Fluc++319901317组BLI信号强度依次为(1.60±0.11)×10^3、(0.934-0.08)×10^5、(0.554-0.07)×10^5和(0.274-0.05)×10^5P·S^-1·CIll^-2·sr^-1,MI+AD-MSC^Fluc+组的BLI信号强度依次为(1.284-0.08)×10^5、(0.56±0.07)×10^5、(0.25±0.05)×10^5和(0.054-0.03)×10^5 P·s^-1·cm^-2·sr^-1,MI+AD-MSC^Fluc+。+39901317组BLI信号强度明显强于MI+AD-MSC^Fluc+组相同时间点(P均〈0.05)。小动物超声检测结果显示术后第28天,MI+PBS组、MI+AD-MSC^Fluc+组和MI+AD-MSC^Fluc++T0901317组小鼠LVEF依次为(27.874-1.81)%、(30.334-1.06)%和(40.494-1.31)%,FS依次为(16.544-0.83)%、(17.83±1.01)%和(25.24±0.99)%,MI+AD-MSC^Fluc++T0901317组小鼠LVEF和Fs均明显高于其他两组(JP均〈0.05)。结论LXR激动NT0901317可提高移植到小鼠梗死心肌的AD-MSC^Fluc+的存活率,并能明显改善心�Objective To investigate the effects of liver X receptor (LXR) agonist on adipose- derived mesenchymal stem cells ( AD-MSCs ) implantation into infarcted hearts of mice. Method AD- MSCr^uc+ which stably expressed firefly luciferase (Fluc) were isolated from β-actin-Fluc transgenic mice and characterized by flowcytometi7. Male FVB mice were randomly allocated into the following four groups (n=10 each): (1) sham group; (2) MI + PBSgroup; (3) MI + AD-MSCnu+ group; (4) MI + ADMSC^Fluc+ + LXR agonist (T0901317) group. AD-MSC^Fluc+ or PBS were injected intramyoeardially into peri- infarcted region of mice heart after permanent left anterior descending ( LAD ) artery ligation. Bioluminensence imaging (BLI)was performed for quantification of injected cells relention and survival. Cardiac function was evaluated by echocardiography.
