摘  要:目的了解妊娠期糖尿病孕妇肝细胞核因子1a外显子7基因变异与血浆胰岛素水平的相关性。方法分析妊娠期糖尿病与正常人的肝细胞核因子1a外显子7基因变异,测定相应血浆胰岛素水平,统计分析它们之间的关系。结果妊娠期糖尿病孕妇肝细胞核因子1a第7号外显子中密码子459的CTG变为CTA,CTG变为TTG,密码子487位AGC变为AAC。这变化为一种多态性。多态性纯合子的血浆胰岛素水平偏低。结论肝细胞核因子1a外显子7基因多态性纯合子孕妇胰岛素分泌水平偏低,提示HNF-1a外显子7基因的多态性改变可能与妊娠期糖尿病的发病有关系。Objective To observe the relativity of hepatic nuclear factor 1 a gene exon 7th polymorphism and plasma insulin concentration in GDM (gestational diabetes mellitus) patients. Methods With PCR and DNA sequen- cing, the genomie DNAs from the blood of GDM patients and normal control were analyzed to find the changes in HNF la gene exon 7th. The changes were added up in groups and compared with plasma insulin concentration. Results In hepatic nuclear factor 1 a gene exon 7th of GDB, Code 459 CTG changed to CTA, CTG changed to TYG, 487 AGC changed to AAC in the 7th exon. The changes were of one kind of polymorphism. I-Iomozygotes of the polymorphism in the group of GDM had lower plasma insulin concentration. Conclusion The polymorphism was found might lead to insulin secretion decrease. It might play a role in the pathogenesis of gestational diabetes mellitus.
