摘 要:目的:探讨丹龙醒脑片对老龄大鼠脑缺血再灌注后NF-κB、ICAM-1表达的影响。方法:用大脑中动脉栓塞再通法建立脑缺血再灌注模型,将动物随机分为假手术组、模型组、丹龙醒脑片组、尼莫地平组,采用原位末端标记法(TUNEL)检测凋亡神经细胞,采用免疫组化法,观察NF-κB、ICAM-1蛋白的表达。结果:丹龙醒脑片能显著减轻神经细胞的损伤;假手术组有一定量的NF-κB、ICAM-1表达,缺血后NF-κB、ICAM-1迅速增加,丹龙醒脑片组能显著抑制NF-κB、ICAM-1的表达,模型组和丹龙醒脑片组比较差异有统计学意义(P<0.01,P<0.05)。结论:抑制由缺血再灌注损伤诱导的NF-κB、ICAM-1表达可能是丹龙醒脑片脑保护作用的机制之一。Objective: To investigate the effect of Danlong Xingnao tablets on NF-κB and ICAM-1 expression in elderly rats after cerebral ischemia/reperfusion.Methods: Rats were randomly divided into sham-operation group,model group,Danlong Xingnao tablet group,and nimodipine group.An animal model of cerebral ischemia/reperfusion was established by middle cerebral artery occlusion-recanalization.The apoptosis of nerve cells was evaluated by terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL).The NF-κB and ICAM-1 expression was measured by immunohistochemistry.Results: Danlong Xingnao tablets could reduce damage to nerve cells.The sham-operation group showed certain NF-κB and ICAM-1 expression,which was rapidly increased after ischemia.The Danlong Xingnao tablet group showed significantly inhibited NF-κB and ICAM-1 expression,with a significant difference between the model group and Danlong Xingnao tablet group(P〈0.01,P〈0.05).Conclusion: The brain-protecting effect of Danlong Xingnao tablets is achieved possibly by inhibiting NF-κB and ICAM-1 expression induced by ischemia/reperfusion injury.
