摘 要:目的评价黄芩水提物中黄酮类化合物对耐甲氧西林的金黄色葡萄球菌(MRSA)抑菌效果。方法采用高效液相色谱分析法检测黄芩水提物中主要黄酮类化合物含量;体外配置与水提物含量相同的各主要黄酮类物质和其混合物;采用微量半固体琼脂稀释法检测各成分对30株MRsA的MIC值;联合抑茵试验采用棋盘稀释法检测各组分间的FIC值,分析其相互作用关系。结果①微量半固体琼脂稀释法检测黄芩苷对30株MRSA的MIC值为1.39mg/ml5.56mg/ml,黄芩素对MRSA的MIC值为0.145mg/ml;黄芩素作用强于黄芩苷,差异具有统计学意义(P〈0.05);汉黄芩苷和汉黄芩素均无抑菌作用。②体外配置四种黄酮类化合物的混合物,混合物对MRSA的MIC稀释比例为1/8-1/16,水提物对MRSA的MIC稀释比例为1/16~1/32,水提物作用强于混合物差异具有统计学意义(P〈O.OS)。③黄芩苷与黄芩素对MRSA的FIC值为3~4,具有相互拮抗作用。结论黄芩水提物中黄芩苷和黄芩素对MRsA有抑菌作用,且黄芩素作用强于黄芩苷;黄芩苷和黄芩素间为相互拮抗作用;汉黄芩苷和汉黄芩素无抑菌作用。Objective To evaluate the inhibitory activity of the main components of water-phase flavonoid extracts from scute- llaria root to methicillin-resistant Staphylococcus aureus(MRSA). Methods Single component suspensions of baicalin, ba- icalein,wogonoside and wagonin were respectively prepared in a concentration as in the water-phase extracts of scutellaria root identified and detected by HPLC. The semisolid agar micro-dilution and chessboard titration were employed to detect the MICs of the suspensions and the inhibitory effect of combinations between the ingredients respectively against 30 clinical isolates of MRSA. Results The MICs of baicalin and baicalein were 1.39 mg/ml to 5.56 mg/ml and 0. 145 mg/ml respec- tively and significantly different (P〈0. 05). No inhibitory activity was found of wogonoside and wagonin against MRSA. A antagonistic action was found between the baicalin and baicalein combination,with the FIC values ranging from 3 to 4. Con- clusion Baicalin and baicalein,with a stronger activity than baicalin,were both shown inhibitory activity against MRSA,although the combined impact of them was antagoniste.
