摘  要:目的采用序列特异性引物聚合酶链反应(PCR-SSP)检测徐州地区汉族人群血小板特异性抗原(HPA)基因多态性。方法2010年期间对徐州地区100名无血缘关系的汉族成年人进行HPA1~17基因及新等位基因Caba+分型研究。根据HPA基因分型试剂盒的要求,调整DNA模板浓度;根据人类生长激素基因(HGH)的保守片段设计内参引物进行PCR—SSP,在包含引物混合液的96孔反应板中,按照相同的循环条件扩增PCR产物;用矿检验比较基因分布期望值与观察值,以验证是否符合Hardy-Weinberg遗传平衡。结果在徐州地区汉族人群的HPA1~17系统中,呈多态性分布的等住基因是HPAla,HPA2a,HPA3a,HPA4a,HPA5a,HPA6a和HPAl5a,其频率分别为0.9950,0.9350,0.5900,0.9950,0.9800,0.9750和0.5750,HPA7~14,16~17及新等位基因Caba+呈单线性分布。结论徐州地区汉族人群HPA的基因有地区特点,人类血小板抗原HPA的基因具有民族多态性。Objective To sequence specific primers polymerase chain reaction (PCR-SSP) detection of Han nationality in Xuzhou area of Human Platelet Antigen (HPA) genetic polymorphism. Methods Studsed on the HPA1-17 gene and allele Caba+ classification 100 unrelated Han Adults. According to HPA genotyping kit requirements during 2010 in Xuzhou. Adjust the DNA template concentration^according to human growth hormone gene (HGH) conservative fragment design inter- nal primers for PCR-SSP, including primers in mixed liquid of 96 hole reaction plate, according to the same loop conditionam- plified PCR product. Used the Xz test comparison of gene distribution expectation value and observation value,to verify com- pliance with Hardy-Weinberg genetic equilibrium. Results In the Han population of Xuzhou region of HPA1-17 system,a polymorphism distribution of alleles were HPAla, HPA2a, HPA3a, HPA4a, HPASa, HPA6a and HPA15a. Its frequency was 0. 995 0,0. 935 0,0. 590 0,0. 995 0,0. 980 0,0. 975 0 and 0. 575 0 respectively. HPA7-14,16-17 and the new allele Caba+ had a single linear distribution. Conclusion HPA gene have regional characteristics in Han people of Xuzhou,human platelet antigen gene have ethnic polymorphism.
