摘  要:目的:研究美托洛尔对大鼠心肌梗死后心力衰竭的保护作用及其可能机制。方法:结扎大鼠左冠状动脉制备心力衰竭大鼠模型,将大鼠随机分为伪手术组、模型组和美托洛尔(10 mg.kg-1.d-1)处理组,并给药6周。6周后进行血流动力学检查,观测心肌细胞凋亡情况以及凋亡相关因子Bcl-2、Bax的表达。结果:美托洛尔能明显改善心功能;减少心肌细胞凋亡;抑制Bcl-2表达,促进Bax表达。结论:美托洛尔对心肌梗死后心力衰竭具有一定的保护作用,其部分机制可能是通过抑制心肌细胞凋亡实现的。Objective: To study the protective effect of metoprolol on heart failure in rats after myocardial infarction. Methods: Heart failure model after myocardial infarction was made by ligating the left anterior descending coronary artery. The rats were randomly divided into sham, model and metoprolol (10rag . kg -1. d -1 ) treated groups. The drug or vehicle was given by gastric perfusion for 6 weeks after operation and then the hemodynamic parameters were detected, TdT-mediated dUTP nick end labe ling was used to examine apoptosis of heart tissues, and western blotting was used to analyze the protein expression of Bcl-2 and Bax. Results: Metoprolol was found to decrease the LVEDP and increase the LVSP and ±LVdp/dt ; compared with model group, metoprolol improved the heart function significantly. Furthermore, metoprolol inhibited myocardial apoptosis which was approved by TUNEL. Additionally, metoprolol inhibited Bax protein expression and promoted Bcl-2 protein expression.Conclusion.. Metoprolol protects the development of heart failure in rats after myocardial infarction probably by inhibiting myocardial apoptosis.
