摘 要:目的:探讨应用丝素-壳聚糖混合膜(silk fibroin-chitosan blend,SFCS)导管移植修复周围神经缺损的效果。方法:选取健康成年雄性日本大耳兔21只,切断双侧面神经颊支,制成8mm的兔面神经颊支缺损模型。使用丝素-壳聚糖混合膜神经导以修复兔右侧面神经缺损,作为实验组;左侧用翻转自体神经修复,作为对照组。术后4、6、8周行神经电生理检查,2、4、6、8周显微镜下观察大体形态,取材行HE及甲苯胺蓝染色组织学检查,并应用图像分析系统进行图像分析。结果:SFCS导管修复组的神经传导速度达到(27.50±3.00)m/s,单位面积(5μm2)的轴突计数为(741.80±49.92),髓鞘厚度达到(12.71±0.42)pm,与自体神经修复组比较差异没有统计学意义(P>0.05)。结论:SFCS神经导管具有促进神经轴突再生的作用,有望成为自体神经移植的替代材料应用于周围神经缺损的修复。Objective: To investigate the effects of SFCS nerve conduits on repairing facial nerve defect in rabbits by transplantation. Methods: Twenty-one healthy male adult rabbits were made into models with 8 mm gaps on both sides of buccal branches of the facial nerve. SFCS nerve conduits were used to repair the right gaps of facial nerve, regarded as the experimental group. Autologous nerve were grafted into the left gaps, regarded as the control group. Electrophysilogical examination of nervous system was performed 4, 6, 8 weeks after operation, and general observation was done with microscope 2,4,6,8 weeks after operation. HE and Toluidine blue staining histological examination were performed, and image analysis system was used to analyze images. Results:The conduction velocity of the facial nerve C (27.50 ±3.00) m/s ] , the counts of axon per unit area ( 741.80 ± 49.92) , and the thickness of the myelin sheath ( ( 12.71 ± 0.42) pm 3 of rabbits in experimental group were not statistically different with those in control group(P 〉 0.05 ). Conclusions : The SFCS nerve conduits can effectively promote axon regeneration of rabbit facial nerve. They can potentially be used as a substitute for autografts to repair nerve defects.
