摘 要:目的探讨丝氨酸-苏氨酸蛋白激酶B(PKB)在糖尿病大鼠骨骼肌中的表达及其与炎性细胞因子肿瘤坏死因子仪(TNF-α)、白细胞介素6(IL-6)的关系。方法将清洁液Wistar大鼠完全随机分为实验组与对照组,各8只。实验组给予高脂、高糖饮食8周后腹腔内注射小剂量链脲佐菌素(30mg/kg),对照组喂养标准大鼠饲料。第13周末处死大鼠,取各组大鼠股四头肌用免疫组织化学方法检测PKB、TNF-α及IL-6的蛋白表达,并分析PKB表达与TNFα、IL-6表达的相关性。结果TNF-α在对照组骨骼肌上表达很少,几乎为0;实验组阳性细胞率为(3.8±1.4)%,2组比较差异有统计学意义(P〈0.05)。IL-6在对照组大鼠骨骼肌上表达率为(2.0±0.8)%;实验组阳性细胞率(5.8±0.7)%,与对照组比较差异有统计学意义(P〈0.05)。PKB在对照组大鼠骨骼肌上阳性率为(9.0±0.9)%;实验组PKB表达减少,阳性细胞率为(2.8±0.8)%,与对照组比较表达明显减少(P〈0.05)。TNF-α、IL-6表达与PKB的表达呈负相关(r=-0.776,r=-0.698,P〈0.05)。结论PKB信号传导系统可以抑制糖尿病大鼠骨骼肌中TNF-α和IL-6的释放。Objective To evaluate the changes of serine/threonine protein kinase B ( PKB), tumor necrosis factor-α(TNF-α) and interleukin 6 (IL-6) expression in quadrieeps femoris muscle of diabetic rats, and to explore the relationship among them. Methods Adult Wistar rats were divided into experimental group ( n = 8 ) and control group ( n = 8 ). Diabetic rats in experimental group were fed by high lipoid and glucose diet. After 8 weeks, they were made by a single injection of streptozotocin (STZ) at a dose of 30 mg/kg. After 13 weeks, they were killed for detecting PKB, TNF-α and IL-6 by quantitative immunohistochemistry assay. Correlation analysis was used to evaluate the relationship among them. Results The expression of TNF -α in the control group in skeletal muscle was almost 0 ; the positive cell rates in the experimental group was ( 3.8 ± 1.4 ) % and there was a significant difference between two groups ( P 〈 0. 05 ). The expression of IL-6 in normal rat skeletal muscle was ( 2. 0 ± 0. 8 ) % ; the positive cell rate in the experimental group was (5.8 -± 0. 7 ) % , which showed a significant difference compared with the control group ( P 〈 0. 05 ). The positive rate of PKB in skeletal muscle of control rats was (9. 0 ± 0. 9)%; the positive cell rate in the experimental group was (2. 8 ± 0. 8 )%, which significantly decreased compared with the control group (P 〈 0. 05 ). There was a significant correlation among PKB and TNF-α, IL-6 ( r = - 0. 776, r = - 0. 698, P 〈 0. 05 ). Conclusion PKB pathway can inhibit the expression of the TNF-α and IL-6 in quadriceps femoris muscle of diabetic rats.
