高效液相色谱法测定头孢克肟颗粒的溶出度

摘　　要：目的采用HPLC法测定头孢克肟颗粒的溶出度。方法色谱柱：Agilent C18（4.6×150mm,5μm）柱,流动相为四丁基氢氧化铵溶液（取10%四丁基氢氧化铵25mL,加水1000mL,摇匀,用1.5mol.L-1磷酸溶液调节pH至7.0-乙腈（72∶28）,检测波长254nm。结果头孢克肟在2.5~15μg.mL-1范围内线性关系良好,r=0.99995（n=6）。结论该方法简便、快速、准确可靠,可以作为头孢克肟颗粒溶出度的测定方法。OBJECTIVE To establish a method of HPLC to determine the dissolution of cefixime granules.METHODS The determination was performed on C18（4.6×150mm,5μm）column,Mobile phase was tetrabutylammonium hydroxide（add 25mL 10% tetrabutylammonium hydroxide to 1000mL water,shake up,adjust to pH 7.2 by 1.5mol·L-1 phosphoric acid）-acetonitrile（72∶28）.The detective wavelength was 254nm.RESULTS The calibration curve was linear in the concentration range of 2.5～15μg·mL-1,r=0.99995（n=6）.CONCLUSION The method is simple,fast and accurate,and could be used for the quality control of the dissolution of cefixime granules.
