摘  要:目的从血脑屏障的角度探讨三七总皂苷对铝暴露脑微血管内皮细胞胞质紧密黏连蛋白1(zonulaoccludens-1,ZO-1)表达的影响,初步揭示三七总皂苷抗老年痴呆的可能靶点及机制。方法采用脑微血管内皮细胞系Balb/c,以含20%胎牛血清和100 mg/L生长因子的DMEM培养基培养,随机分为正常组、模型组、铝螯合剂组及三七总皂苷低、中、高剂量组,经铝暴露24 h后,分别以荧光实时定量PCR和Western blot的方法测定ZO-1的基因转录和蛋白表达水平,观察不同剂量三七总皂苷对ZO-1表达的影响。结果与正常组相比,铝暴露24 h可使脑微血管内皮细胞ZO-1的转录及表达明显减少(P<0.01)。与模型组比较,三七总皂苷高剂量组能显著增加ZO-1的基因转录水平(P<0.05),中、高剂量组能显著提高ZO-1的蛋白表达(P<0.05,P<0.01)。结论三七总皂苷对铝暴露下的脑微血管内皮细胞具有一定的保护作用,可增加血脑屏障紧密连接主要组成蛋白ZO-1的表达。Objective From the perspective of blood-brain barrier (BBB), to study the influence of panax notoginseng saponins (PNS) on tight junctional protein zonula occludens-1 (ZO-1) in BBB in vitro exposed to aluminum, in order to preliminary explore the possible target and mechanism of PNS against Alzheimer's disease (AID). Methods The BBB in vitro was established by culturing mouse brain microvascular endothelial cells line Balb/c which cultured in DMEM medium containing 20% fetal bovine serum and 100 mg/L growth factor. The cells were randomly divided into normal control group, model group, aluminum chelating agent group and low-, medium- and high-dose of PNS group. The injured model of brain microvascular endothelial cells was simulated by exposing to aluminum for 24 h. The gene transcription and protein expression of ZO-1 were measured by fluorescence real-time quantitative PCR and Western blot respectively. Meanwhile, the changes of expression of ZO-1 caused by different dose of PNS were detected. Results Compared with control group, the transcription and expression of ZO-1 in brain microvascular endothelial cells was reduced significantly after exposure to aluminum for 24 h (P〈0.05). PNS could increase the gene transcription and protien expression of ZO-1 in endothelial ceUs injured by aluminum. Compared with model group, there was statistical significance in level of gene transcription in medium-dose group (P 〈0.05) and level of protien expression in both medium-and high-dose group (P 〈0.05, P 〈0.01). Conclusion PNS has protective effect on brain microvascular endothelial cells and could improve the expression of main component protein ZO- 1 of BBB exposed to aluminum.
