补肾清热方对流产大鼠子宫蜕膜组织侵袭相关因素调控的影响

摘　　要：目的研究补肾清热方对于溴隐亭致大鼠流产模型的治疗作用及其机理。方法将受孕大鼠分为空白对照组、模型组、黄体酮组及中药高、低剂量组,皮下注射溴隐亭造模,采用补肾清热方进行治疗,以黄体酮作为阳性对照药观察,观察胎仔数,计算流产率,放射免疫法检测大鼠血清雌二醇(E2)、孕酮(P),免疫组化法检测大鼠子宫蜕膜组织基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶组织抑制物-3(TIMP-3)含量。结果与模型组比较,各治疗组大鼠子宫内胎仔数及流产率差异有统计学意义(P<0.01),中药高剂量组大鼠血清雌激素升高、孕激素上升,子宫蜕膜组织MMP-9表达增加、TIMP-3表达下调,差异均有统计学意义(P<0.05,P<0.01)。结论补肾清热方具有保胎作用,并对子宫蜕膜组织浸润相关因素具有调节平衡作用。Objective To research the therapeutic action and mechanism of Bushenqingre Fang in treating abortion rats model induced by bromocriptine. Methods The pregnant rats were divided into blank control, model group, progestin group, herbal medicine high-dose group and herbal medicine low-dose group. Rats were subcutaneously injected bromocriptine to make abortion model and then treated with Bushenqingre Fang. With progestin as the positive control, the number of fetus was observed to calculate the abortion rate. Radiation immune method was used to detect the serum E2, P, and immunohistochemical method to determine MMP-9, TIMP-3 of uterine decidual tissue. Results Compared with model group, fetus number and abortion rate of the treatment groups were significantly different （P 〈 0.01）. In high dose of herbal medicine group, serum estrogen obviously elevated, progesterone raised, the expression of MMP-9 increased and TIMP-3 decreased in uterine decidual tissue （P〈0.05, P〈0.01）. Conclusion Bushenqingre Fang can regulate the balance of the relative factors invading uterine decidual tissue, and protect fetus.
