朱砂七抗单纯疱疹病毒Ⅰ型的体外实验研究

摘　　要：目的研究朱砂七粗提物和总蒽醌体外对单纯疱疹病毒Ⅰ型(HSV-Ⅰ)的抑制作用。方法运用HSV-Ⅰ,以非洲绿猴肾细胞(Vero)作为宿主细胞,通过三种给药方式观察感染病毒后的细胞变性反应(CPE),MTI比色法检测不同浓度朱砂七粗提物和总蒽醌对HSV-Ⅰ的直接杀灭、预防和阻止复制作用。结果朱砂七粗提物和总蒽醌能抑制HSV-Ⅰ病毒的复制,但无预防和直接杀灭作用,朱砂七粗提物抑制HSV-Ⅰ病毒复制IC50为0.031 5g/L,治疗指数(TI)为19.84;总蒽醌抑制HSV-Ⅰ病毒复制IC50为0.004 7 g/L,治疗指数(TI)为16.62。结论朱砂七粗提物和总蒽醌在体外具有明显的保护宿主细胞抵抗HSV-Ⅰ病毒复制的作用。Objective It is to study the antiviral effects of Zhushaqi crude extractions and total anthraquinone on herpes simplex virus type I （ HSV - I ） in vitro. Methods HSV - I was used to infect vero cell that used as host cell. Cells degen- eration reactions of three kinds of administration were observed after virus infection. The killing effect directly on virus, virus re- production inhibition and prevention of different dosage of Zhushaqi crude extracts and total anthraquinone on HSV - I were observed by MTT. Results Zhushaqi could lessen the cytopathie effect induced by HSV - I and inhibit virus replication, but could not prevent and kill virus directly. The ICs0 of the Zhashaqi crude extractings and total anthraquinone against HSV - I were 0. 031 5 g/L, 0. 004 7 g/L respectively. The anti-virus therapeutic index （TI） were 19.84,16.62. Conclusion Zhushaqi crude extracts and total anthraquinone showed significant activity of protecting host cells and antiviral effect on HSV - I in vitro.
