摘  要:目的:观察益气活血化浊中药对代谢综合征(MS)模型大鼠的干预作用。方法:雄性SD大鼠60只,高脂高盐饲料喂养复制MS大鼠模型。随机分为正常组、模型组、文迪雅组、及中药(低、中、高剂量)组,中药组予中药、正常组和模型组予等体积0.9%氯化钠溶液灌胃;治疗4周后测血糖、胰岛素、血脂、瘦素、胰岛素敏感指数(ISI)、脂肪组织过氧化物增殖体激活受体(PPARγmRNA)表达。结果:①中药各组体质量、内脏脂肪重量及系数、瘦素水平均显著低于模型组(P<0.05,P<0.01),但文迪雅组降低无统计学意义;②中药中剂量和文迪雅组的餐后2h血糖、血脂均显著性低于模型组而ISI显著高于模型组(P<0.05);③中药中剂量组PPARγmRNA表达比值显著高于模型组(P<0.05),但低于文迪雅组(P<0.01)。结论:该复方中药促进MS大鼠脂肪组织PPARγmRNA表达作用虽弱于文迪雅,但兼有纠正糖脂代谢紊乱、减肥、改善胰岛素、瘦素抵抗等多重作用。Objective: To observe the infulence of compound TCM with strengthening Qi, promoting blood circulation and discharging trash function on SD rats with metabolic syndrome(MS). Methods: Fed 60 male SD rats with high fat-salt diet to establish a model which resembled human character of MS. Rats were randomly divided into groups: normal group, model group, rosiglitazone group, low-dose, middle-dose and high-dose TCM groups. The TCM groups were administered intragastrically with compound TCM, normal and model group with equal volume normal saline. Administered for 4 weeks, tested blood glucose, insulin, serum lipid and leptin, detected PPAR γ expression of omental grease by RT-PCR. Results: ①In compound TCM groups, body weight, weight and coefficient of visceral fat were significantly lower than those in model group(P〈0.05, P〈0.01) and the decrease in rosiglitazone group had no statistical significance. ②In rosiglitazone and middle-dose TCM group, the blood glucose and serum lipid were significantly lower and ISI was higher than those in model group(P〈0.05). ③In middle-dose TCM group, expression of PPAR γ mRNA was significantly higher(P〈0.05) than in model group, was lower than in rosiglitazone group(P〈0.01). Conclusion: These indicate that the compound TCM with strengthening Qi, promoting blood circulation and discharging trash function has an effect on promoting omental grease PPAR γ mRNA expression, which is weaker than rosiglitazone and has combining multiple effects of enhancing insulin sensitivity, retrieving sugar-lipid metabolic disorder, decreasing body weight and improving leptin resistance.
