摘  要:目的:研究南方红豆杉种子萌发的机制,即南方红豆杉种子形态后熟过程中的生物大分子的变化过程。方法:采用0.05%赤霉素(GA3)加低(4℃)-暖(23℃)-低(4℃)变温层积处理南方红豆杉种子,研究其生理生化变化。结果:采用激素加变温层积法处理种子后,蛋白质、氨基酸、可溶性糖都具有在低温层积时期累积较快,暖层积时期消耗较快的特点,三者的含量总体都呈上升趋势,分别比处理前增加115.56%、123.70%、50.71%,但脂肪含量下降40.00%。脱氢酶、过氧化物酶(POD)、多酚氧化酶(PPO)、蛋白酶活性分别比处理前增加111.05%、178.73%、141.84%、169.97%。结论:激素加变温层积法影响南方红豆杉种子生理生化变化,使种子内贮藏物质不断分解代谢、转化和利用,这也是破除其休眠的关键。Objective: Further study the mechanism of germination of Taxus mairei (Lemee et Levi.) S.Y.Hu ex Liu seeds, that is the change of biological macromolecules during the process of morphological afterripening of Taxus mairei seed. Methods: Using the seeds handled by cold-warm-cold stratification treatments with 0.05% GA3 to break dormancy, to determine the physiological changes. Results: After using the seeds handled by cold(4℃)-warm(23℃)-cold(4℃) stratification treatments with excitatory antacoid, all the contents of protein, amino acid and soluble sugar have the features of faster accumulation during the process of cold stratification and consumed faster during the process of warm stratification. The contents of all are rising overall, increased 115.56%, 123.70%, 50.71% after treatments, respectivlely. But the contents of fat are declined to 40.00%. The activity of dehydrogenase, peroxidase, PPO, protease are increased to 111.05%, 178.73%, 14L84%, 169.97% compared with before treatments. Conclusion: Using cold-warm-cold stratification treatments with excitatory autacoid influnces the physiological activity in seeds. The key to break dormancy of seed maybe is the decomposition, metabolism, transformation and use of reserved substances.
