牦牛多条带型乳酸脱氢酶同工酶的H亚基由杂合型Ldhb基因编码

摘　　要：通过聚丙烯酰胺凝胶电泳在背最长肌匀浆液中检测到呈多条带型的乳酸脱氢酶(LDH)同工酶谱的麦洼牦牛个体,从此个体背最长肌中提取总RNA,采用RT-PCR方法克隆编码LDH的H亚基的Ldhb基因cDNA序列.序列比对分析结果为两种序列,分别对应GenBank中牦牛的Ldhb-F和Ldhb-S的基因序列,证实了这种LDH特殊带型是由Ldhb-F和Ldhb-S杂合型基因编码的.A Maiwa yak individual with multiple stained bands of lactate dehydrogenase （LDH） i：;oenzymes is discovered by polyacrylamide gel electrophoresis of longissimus dorsi homogenate, and the cDNA sequence ofLdhb gene encoding H subunit of LDH is obtained by RT-PCR using total RNA from Iongissimus dorsi of this yak. Sequence analysis reveals two sequences corresponding to the sequences of Ldhb-F and Ldhb-S in GenBank respectively, demonstrating that the special stained bands are encoded by heterozygous Ldhb gene （Ldhb-F and Ldhb-S）.
