摘 要:目的:观察电纺丝可降解聚氨酯材料(PU)的生物相容性。方法:利用小鼠L-929细胞和人类脐静脉内皮细胞(HUVEC),对PU进行细胞毒性试验和贴附增值试验,观察其细胞毒性及其对实验细胞的贴附增值性能的影响。细胞毒实验中共分为三组PU/BDO组,PU/LYS组,对照组;细胞贴附实验中共分为三组PU/BDO组,PU/LYS组,PTFE组。结果:细胞毒实验中各组的细胞毒性评级均为0或1级;在电镜下观察,细胞贴附实验中种植HUVEC时,在PU/BDO组或PU/LYS组,细胞呈单层融合贴附增殖,PTFE组呈单层分散贴附增殖;种植L-929细胞时,在PU/BDO组或PU/LYS组,细胞呈多层融合贴附增殖,PTFE组呈多层分散贴附增殖。结论:PU有较低的细胞毒性;和PTFE材料相比,更易使L-929细胞或HUVEC细胞贴附增值。Objective:To explore the biocompatibility of electrospinning polyurethane(PU).Methods: The evaluation on biocompatibility of electrospinning PU was performed by rat L-929 fibroblasts(l-929) or human umbilical vein endothelial cells(HUVECs) for cytotoxicity and cell adhesion /proliferation tests.In the cytotoxic test,the test was done on three groups:the group PU/BDO,group PU/LYS and the control group,while in the cell adhesion /proliferation tests,he test was carried out on three groups:the group PU/BDO,group PU/LYS,and the PTFE(control).Results: The levels of toxicity in each group was level 0 or 1.Observed by SEM,the L-929 and HUVEC cells could attach and proliferate on the electrospinning PU materials.L-929 cells were overlapped on the electrospinning PU/BDO or PU/LYS,and HUVEC were monolayer confluence on the surfaces of them.However,on the surfaces of PTFE,L-929 showed multiple disperse overlapping,while HUVEC performed monolayer confluence.Conclusion: The cytotoxicity of electrospinning PU is low;compared to PTFE,the L929 and HUVEC cells could attach and proliferate on it more easily.
