中医不同方法对骨质疏松症大鼠骨骼、骨骼肌Ⅰ型胶原蛋白表达影响的实验研究

摘　　要：目的观察骨质疏松症大鼠骨骼、骨骼肌Ⅰ型胶原蛋白表达变化的影响,探讨中医防治骨质疏松症的作用机制。方法将120只雌雄各半的大鼠随机分为正常对照组、模型对照组、补肾中药组、健脾中药组、活血化瘀中药组和骨疏康中药组6个组。采用Western blot方法测定大鼠骨骼、骨骼肌Ⅰ型胶原蛋白表达。结果①各组大鼠骨组织均可检测到Ⅰ型胶原的蛋白表达。与正常组比较,各组大鼠骨组织Ⅰ型胶原的蛋白表达均明显下降(P<0.01);与模空组比较,各治疗组大鼠骨组织Ⅰ型胶原的蛋白表达均明显升高,其中补肾组、骨疏康组升高趋势最显著(P<0.01)。②各组大鼠骨骼肌均可检测到Ⅰ型胶原的蛋白表达。与正常组比较,各组大鼠骨骼肌Ⅰ型胶原的蛋白表达均明显下降(P<0.01);与模空组比较,各治疗组大鼠骨骼肌Ⅰ型胶原的蛋白表达均明显升高(P<0.01),其中补肾组升高趋势最明显。结论①骨质疏松症的形成,可能与骨骼、骨骼肌Ⅰ型胶原表达的异常变化有关。②补肾、健脾方法通过调控骨质疏松症大鼠的骨骼、骨骼肌Ⅰ型胶原蛋白表达,对骨质疏松症具有一定的防治作用。Objective To explore the mechanism of traditional Chinese herb in treating osteoporosis by investigating protein expression of collagen I （ COL I） in skeleton and skeletal muscle of osteoporotic rats. Methods One hundred and twenty rats （male/female = 1 ： 1 ） were randomly divided into 6 groups as following： blank control group, model control group, invigorating the kidney group, invigorating the spleen group, promoting blood flow group, and Gusukang group. The protein expression of COL I in skeleton and skeletal muscle was tested using Western-blotting. Results 1 ） Protein expression of COL I could be detected in the skeleton of all the groups. Compared to blank control group, the expression in all other groups decreased significantly （P 〈 0. 05）. Compared to model control group, the expression in all the herbal treating groups elevated significantly, among which the expression in invigorating the kidney group and Gusukang group elevated most significantly （P 〈0.01 ）. 2） Protein expression of COL I could be detected in skeleton muscle of all the groups. Compared to blank control group, results in all other groups decreased significantly （ P 〈 0. 05 ）. Compared to model control group, expression in all herbal treating groups elevated significantly, among which the expression in invigorating the kidney group elevated most significantly （P 〈 0. 01 ）. Conclusion 1 ） The development of osteoporosis may be associated with abnormal expression of COL I in the skeleton and skeletal muscle. 2） Treatments of invigorating the kidney and invigorating the spleen show its efficacy to prevent osteoporosis by regulating the protein expression of COL I in the skeleton and skeletal muscle.
