摘  要:目的:观察大鼠脊髓急性损伤早期脊髓内iNOS及细胞凋亡的变化。方法:54只SD大鼠随机分为对照组(6只)和实验组(8个时间点,各6只);实验组采用Allen’s方法制作大鼠脊髓损伤模型。采用免疫组织化学染色观察对照组和实验组脊髓损伤后3h、6h、12h、1d、3d、5d、7d和14d脊髓灰质和白质中iNOS阳性细胞数的变化,采用TUNEL法观察脊髓凋亡细胞的变化,计算凋亡指数。结果:不同组间脊髓灰质和白质iNOS阳性细胞数及凋亡指数差异均有统计学意义(F=197.586、118.380、345.184,P<0.001);脊髓损伤后,灰质和白质内iNOS阳性细胞数持续增加,并于伤后3d达到高峰,持续到伤后7d,伤后14d减少;细胞凋亡于损伤后7d达到高峰,伤后14d减少,但均高于对照组(P<0.05)。结论:iNOS变化和细胞凋亡的关系呈现出的时序性规律可望用于脊髓损伤时间的推断,也为脊髓损伤的诊断及治疗提供了依据。Aim:To observe the changes of both inducible nitric oxide synthase (iNOS) and apoptosis after acute spinal cord injury(SCI) in rats.Methods:A total of 54 SD rats were randomly allocated into control group and 8 experimental groups(6 in each group).The rats in the experimental groups were established SCI model.At 3 h,6 h,12 h,1 d,3 d,5 d,7 d and 14 d after SCI,the iNOS positive cells in white and grey matter of spinal cord were marked by immunohistochemical staining,and nerve cell apoptosis was tested by TUNEL at each time point.Results:The iNOS positive cell number in white matter and grey matter and apoptosis index among the groups had significant difference(F=118.380,197.586,,345.184,P0.001).The expression level of iNOS reached peak at 3 days after injury,then kept at a high level until 7 days after injury,and then reduced significantly after 14 days. The apoptosis index reached peak at 7 days after injury,then reduced significantly after 14 days.Conclusion:Time-dependent changes of both iNOS and cell apoptosis could be used for both diagnosis and treatment of SCI,and estimation of the time of SCI in forensic pathology.
